益肺活血湯對(duì)大鼠慢性阻塞性肺疾病模型肺組織氧化抗氧化失衡的影響

【摘要】:[目的]觀察益肺活血湯對(duì)大鼠慢性阻塞性肺疾病(COPD)模型病理和肺功能的影響,評(píng)價(jià)其在氧化/抗氧化失衡機(jī)制方面的干預(yù)效應(yīng)。[方法]采用SPF級(jí)雄性SD大鼠40只隨機(jī)分為對(duì)照組10只、COPD組15只、益肺活血湯組15只。COPD組采用氣管內(nèi)注射脂多糖聯(lián)合香煙煙霧吸入法制備大鼠COPD模型同時(shí)給予生理鹽水灌胃。益肺活血湯組在COPD組制備方法基礎(chǔ)上同時(shí)給予益肺活血湯灌胃治療。對(duì)照組則采用室內(nèi)環(huán)境喂養(yǎng),氣管內(nèi)注射液生理鹽水,同時(shí)給予生理鹽水灌胃。模型制備及治療過(guò)程結(jié)束后檢測(cè)各組大鼠肺功能,并檢測(cè)肺組織中谷胱甘肽(GSH)、丙二醛(MDA)和超氧化物歧化酶(SOD)含量,轉(zhuǎn)錄因子紅細(xì)胞系-2p45(NF-E2)相關(guān)因子-2(Nrf2)和γ-谷氨酰半胱氨酸合成酶(γ-GCS)蛋白及m RNA表達(dá)。[結(jié)果]與對(duì)照組比較,模型組肺功能及肺組織病理切片提示COPD模型制備成功,MDA、GSH增高,SOD下降,NRF2和γ-GCS的蛋白及m RNA表達(dá)增高。與模型組比較,治療組肺功能和肺組織病理變化減輕,MDA、GSH下降,SOD增高,NRF2和γ-GCS的蛋白及m RNA表達(dá)下降。[結(jié)論]益肺活血湯能有效減輕氣管內(nèi)注射脂多糖聯(lián)合香煙煙霧吸入所造成的大鼠肺功能下降和肺組織病理變化。其機(jī)制應(yīng)與通過(guò)提高肺組織SOD的含量從而減輕氧化應(yīng)激有關(guān),與激活Keap1-Nrf2-ARE通路引起γ-GCS基因和蛋白水平上調(diào)、GSH增多相關(guān)可能性小。
[Abstract]:[objective] to observe the effect of Yifei Huoxue decoction on pathology and pulmonary function of chronic obstructive pulmonary disease (COPD) model in rats. [methods] Forty male SD rats of SPF grade were randomly divided into control group (n = 10) and control group (n = 15). COPD model of rats was established by intratracheal injection of lipopolysaccharide combined with cigarette smoke inhalation. The group of Yifei Huoxue decoction was given intragastric administration of normal saline. The group of Yifei Huoxue decoction was treated with Yifei Huoxue decoction on the basis of the preparation method of COPD group. The control group was fed with indoor environment. Rats were given intratracheal injection of normal saline at the same time. The lung function of each group was measured after the preparation and treatment of the model, and the contents of glutathione glutathione (GSH), malondialdehyde (MDA) and superoxide dismutase (SOD) in lung tissue were detected. The protein and m RNA expression of transcription factor erythrocyte line -2p45NF-E2) and 緯 -glutamyl cysteine synthase (緯 -GCSase) were compared with those of control group. The lung function and pathological section of lung tissue in the model group suggested that the COPD model was successfully prepared by increasing the GSH and decreasing the expression of protein and m RNA of NRF2 and 緯 -GCS, compared with the model group. In the treatment group, lung function and pathological changes of lung tissue reduced the expression of NRF2 and 緯 -GCS protein and m RNA. [conclusion] Yifei Huoxue decoction can effectively reduce the large amount caused by endotracheal injection of lipopolysaccharide combined with cigarette smoke inhalation. The mechanism of the decrease of lung function and the pathological changes of lung tissue may be related to reducing oxidative stress by increasing the content of SOD in lung tissue. There is little correlation between activation of Keap1-Nrf2-ARE pathway and up-regulation of 緯 -GCS gene and protein level.
