外源性腦源性神經(jīng)因子對遺傳性聾動物模型耳蝸形態(tài)和功能的影響

【摘要】:目的:研究外源性腦源性神經(jīng)因子(BDNF)對條件敲除縫隙鏈接蛋白26(Cx26)小鼠耳蝸形態(tài)和功能的影響。方法:選取條件敲除的Cx26小鼠為動物模型,在出生后2d(P0-P2)通過中階注射給予外源性BDNF。分別觀察BDNF在耳蝸中的表達情況及其對神經(jīng)纖維生長的促進作用,注射后60d,BDNF對corti器及螺旋神經(jīng)節(jié)細胞的保護作用,并檢測注射后動物聽力的改變情況。結(jié)果:(1)外源性BDNF可在實驗動物體內(nèi)良好表達;(2)外源性BDNF對動物耳蝸神經(jīng)纖維生長有良好的促進作用;(3)外源性BDNF可有效保護corti器的形態(tài);計數(shù)耳蝸各回螺旋神經(jīng)節(jié)細胞數(shù),以未注射耳頂回的密度值作為標準單位,注射耳中回和底回的密度值分別為0.81±0.02、0.65±0.02,與未注射耳相比,均差異有統(tǒng)計學(xué)意義(均P0.05);(4)通過計算注射耳與未注射耳各頻率ABR閾值,顯示兩者差異無統(tǒng)計學(xué)意義。結(jié)論:早期給予外源性BDNF可有效保護條件敲除Cx26小鼠耳蝸的corti器及螺旋神經(jīng)節(jié)細胞的形態(tài),但對于聽功能則無保護作用。
[Abstract]:Objective: to study the effect of exogenous brain-derived neurofactor (BDNF) on cochlea morphology and function in mice with conditional knockout gap link protein 26Cx26. methods: Cx26 mice with conditional knockout were selected as animal models. The expression of BDNF in cochlea and its promoting effect on nerve fiber growth were observed, and the protective effects of BDNF on corti organ and spiral ganglion cells were observed 60 days after injection. Results the exogenous BDNF could well express the growth of cochlear nerve fibers in experimental animals. The exogenous BDNF could promote the growth of cochlear nerve fibers in animals. [results] exogenous BDNF could effectively protect the morphology of the corti organ. The number of spiral ganglion cells in each gyrus of the cochlea was counted. The density of the parietal gyrus was taken as the standard unit. The density values of the middle and bottom gyrus of the injected ears were 0.81 鹵0.02 and 0.65 鹵0.02, respectively, compared with those of the uninjected ears. The difference was statistically significant (P 0.05). The frequency ABR thresholds of injected and uninjected ears were calculated. Conclusion: early administration of exogenous BDNF can effectively protect cochlear corti organ and spiral ganglion cells of Cx26 mice, but have no protective effect on auditory function.
