電腦輻射對(duì)小鼠部分器官組織形態(tài)學(xué)上的影響

【摘要】:本研究把40只4周齡昆明種雄性小白鼠(體重:23.0±2.7g/只)適應(yīng)性飼養(yǎng)一周后,隨機(jī)分為兩組:A組(對(duì)照組)、B組(實(shí)驗(yàn)組)每組20只。對(duì)照組在室內(nèi)光源下正常飼養(yǎng),實(shí)驗(yàn)組將輻射強(qiáng)度為0.31μT筆記本電腦架于鼠籠上面,輻射時(shí)間為每天上午輻射4小時(shí),下午輻射4小時(shí),中午停止輻射1小時(shí),連續(xù)輻射28天后,先用眼眶后靜脈叢采血法采血1~2mL后,取睪丸、腎、肺、部分空腸、氣管等器官,然后用石蠟切片法制作組織切片并進(jìn)行H-E染色;利用顯微數(shù)字圖像分析處理系統(tǒng)觀察組織結(jié)構(gòu)的變化;血樣用沙利氏比色法測(cè)定血紅蛋白含量;用蛋白含量測(cè)量的方法測(cè)定小鼠睪丸中的Caveolin-1蛋白含量。以這些指標(biāo)來(lái)研究和探討電腦輻射對(duì)小鼠部分器官組織形態(tài)學(xué)上的影響,從而探究其對(duì)人體各器官組織形態(tài)學(xué)上的影響及細(xì)胞損傷過(guò)程中Caveolin-1蛋白的表達(dá)變化,為進(jìn)一步研究細(xì)胞損傷機(jī)理提供理論基礎(chǔ)和科學(xué)依據(jù)。 結(jié)果顯示: 1.實(shí)驗(yàn)組的采食量及飲水量明顯(P 0.05)小于對(duì)照組;實(shí)驗(yàn)組的平均體重從輻射2周后開始明顯(P 0.05)小于對(duì)照組的平均體重;實(shí)驗(yàn)組血紅蛋白含量(13.7±0.3g/100mL)明顯(P 0.05)少于對(duì)照組(15.1±0.3g/100mL)血紅蛋白含量。 2.實(shí)驗(yàn)組空腸的小腸絨毛排列不規(guī)則,變疏松,有的變短,甚至出現(xiàn)尖端斷裂脫落等現(xiàn)象,黏膜上皮的吸收細(xì)胞出現(xiàn)萎縮及壞死,且吸收細(xì)胞游離緣上的紋狀緣有脫落現(xiàn)象。 3.實(shí)驗(yàn)組氣管的軟骨萎縮,氣管腔里有很多異物,粘膜上皮細(xì)胞萎縮,甚至伴有脫落的現(xiàn)象。 4.實(shí)驗(yàn)組的細(xì)支氣管,終末細(xì)支氣管及呼吸性細(xì)支氣管的上皮細(xì)胞發(fā)生萎縮現(xiàn)象,排列變疏松,柱狀細(xì)胞上緣的纖毛有脫落。部分肺泡管、肺泡囊、肺泡伴有充血,肺泡隔里的纖維明顯增生,伴有炎性細(xì)胞的浸潤(rùn)。 5.實(shí)驗(yàn)組的生精小管中各級(jí)生精細(xì)胞發(fā)生萎縮,數(shù)量減少,管腔內(nèi)精子數(shù)量減少且很多精子出現(xiàn)畸形現(xiàn)象。睪丸間質(zhì)明顯減少,間質(zhì)細(xì)胞變得模糊有的甚至消失。 6.實(shí)驗(yàn)組腎小球玻璃樣變性,纖維化,有些腎小球代償而肥大,一些腎小管發(fā)生萎縮,有些腎小管代償擴(kuò)張,間質(zhì)結(jié)締組織有增生現(xiàn)象。 7.實(shí)驗(yàn)組睪丸中Caveolin-1蛋白表達(dá)減少。
[Abstract]:In this study, 40 4-week-old Kunming male mice (weight of 23.0 鹵2.7 g / mouse) were fed adaptively for one week, and were randomly divided into two groups: group A (control group, n = 20), group B (control group, n = 20) and control group (n = 20). In the experimental group, the radiation intensity of 0.31 渭 T notebook computer was placed on the cage. The radiation time was 4 hours in the morning, 4 hours in the afternoon, 1 hour at noon, and 28 days after continuous radiation. The testis, kidneys, lungs, part of jejunum, trachea and other organs were collected from the blood collected from the posterior orbital venous plexus at 1: 2 mL, and then the tissue sections were made by paraffin section and stained with H-E. The changes of tissue structure were observed by means of microscopic digital image analysis and processing system, and hemoglobin content was determined by hematocrit colorimetry in blood samples. The content of Caveolin-1 protein in testis of mice was measured by protein content measurement. The effects of computer radiation on the morphology of some organs of mice were studied and discussed by using these indexes. In order to provide theoretical basis and scientific basis for further study on the mechanism of cell injury, the effects of Caveolin-1 on the morphology of human organs and tissues and the expression of Caveolin-1 protein in the process of cell injury were investigated. The results show that:. 1. The intake and water intake of the experimental group were significantly lower than that of the control group (P 0.05), and the average body weight of the experimental group was significantly lower than that of the control group from 2 weeks after radiation. The hemoglobin content in the experimental group was 13.7 鹵0.3g / 100mL) significantly lower than that in the control group (15.1 鹵0.3g / 100mL). 2. In the experimental group, the villi of the jejunum were arranged irregularly and loosened, some of them became shorter, and even the tip was broken off, the resorption cells of the mucosal epithelium were atrophied and necrotized, and the striate edge of the free edge of the resorption cell was shedded. 3. In the experimental group, there were many foreign bodies in the trachea, atrophy of mucosal epithelium and even exfoliation. 4. The epithelial cells of the bronchioles, terminal bronchioles and respiratory bronchioles of the experimental group were atrophied, arranged loosely, and the cilia of the upper edge of the columnar cells shedded. Part of the alveolar duct, alveolar sac, alveolar vesicle, alveolar accompanied by congestion, The fibers in the alveolar septum were proliferated, accompanied by inflammatory cell infiltration. 5. The spermatogenic cells in the seminiferous tubules of the experimental group were atrophied, the number of spermatozoa in the tubules decreased, and many spermatozoa deformed, the interstitial cells of the testis decreased obviously, and some of the interstitial cells became blurred or even disappeared. 6. Glomerular vitreous degeneration, fibrosis, some glomerular compensatory hypertrophy, atrophy of some renal tubules, compensatory dilatation of some tubules and hyperplasia of interstitial connective tissue in the experimental group. 7. The expression of Caveolin-1 protein in testis of experimental group was decreased.
