【摘要】：目的探究坤泰膠囊(黃連、黃芩、阿膠,等)對(duì)去氧乙烯基環(huán)乙烯(VCD)所致卵巢功能不全(POI)模型大鼠的療效。方法 75只21日齡離乳大鼠,隨機(jī)均分為佐劑對(duì)照組,模型組,坤泰膠囊低、中、高劑量組。除佐劑對(duì)照組腹腔注射芝麻油外,其余4組均給予腹腔注射VCD溶液。灌胃給藥45 d。每組選取8只持續(xù)處于動(dòng)情間期或持續(xù)動(dòng)情周期紊亂的大鼠,進(jìn)行檢測(cè)大鼠血清促卵泡生成素(FSH)、促黃體生成素(LH)、雌二醇(E_2)、抗苗勒管激素(AMH);剖腹取出卵巢、子宮稱重并進(jìn)行形態(tài)學(xué)觀察。結(jié)果與佐劑對(duì)照組相比,模型組大鼠血清FSH明顯增高且AMH、E_2顯著降低,LH有所增高;經(jīng)坤泰中、高劑量干預(yù)后,FSH水平顯著降低,AMH水平明顯增加;模型組大鼠卵巢濕重較佐劑對(duì)照組減輕,坤泰膠囊干預(yù)后,低劑量組卵巢濕重增加明顯;模型組大鼠各級(jí)卵泡及黃體計(jì)數(shù)均明顯減少,坤泰膠囊干預(yù)后,各級(jí)卵泡計(jì)數(shù)明顯增加,黃體計(jì)數(shù)有所增加;光鏡下,模型組子宮內(nèi)膜變薄,管壁子宮腺體數(shù)量明顯減少。經(jīng)坤泰膠囊干預(yù)后,內(nèi)膜有所增厚,腺體數(shù)量明顯增加。結(jié)論坤泰膠囊可以減緩模型大鼠卵巢功能衰減的速度。
[Abstract]:Objective to investigate the effect of Kun Tai capsule (Coptis chinensis, Radix Scutellariae, Gelatin, et al.) on ovarian dysfunction induced by deoxyvinyl cycloethylene (VCD) in rats. Methods Seventy-five 21-day-old isolated rats were randomly divided into adjuvant control group and model group. Except adjuvant control group, the other four groups were given intraperitoneal injection of VCD solution. Each group was given intraperitoneal injection of VCD solution for 45 days. Eight rats in each group were continuously in the period of estrus or continuous estrous cycle disorder. The serum follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E _ 2), anti-mullerian hormone (AMHN) were detected in rat serum, ovary was taken out by laparotomy, uterus was weighed and morphological observation was made. Results compared with adjuvant control group, In the model group, the serum FSH level was significantly increased and AMHH E2 was significantly decreased, while the serum FSH level was significantly decreased after high dose intervention, the wet weight of ovary in the model group was less than that in the adjuvant control group, and after the intervention of Kun Tai capsule, the rats in the model group were significantly lower than those in the adjuvant control group, and the rats in the model group were treated with Kuntai capsule. The number of follicles and corpus luteum in model group decreased obviously after intervention of Kun Tai capsule, the count of follicle and corpus luteum was increased, and the endometrium of model group was thinned under light microscope, the wet weight of ovary was increased obviously in low dose group, and the count of follicle and corpus luteum in model group decreased obviously after intervention of Kun Tai capsule. The number of uterine glands in the tube wall decreased obviously. After the intervention of Kuntai capsule, the endometrium was thicker and the number of glands was obviously increased. Conclusion Kun Tai capsule can slow down the decline of ovarian function in the model rats.
