腸道白色念珠菌感染對脾虛小鼠小腸黏膜中T細胞亞群和穿孔素-顆粒酶B的影響

【摘要】:目的:通過觀察腸道白色念珠菌感染對脾虛小鼠小腸黏膜固有層中T細胞亞群和穿孔素、顆粒酶的影響,探討機體在脾虛狀態(tài)下感染白色念珠菌的部分免疫機制。方法:本實驗采用飲食失節(jié)加勞倦過度的復合因素制備小鼠脾虛模型,經(jīng)腸道感染白色念珠菌后第14天,應用流式細胞術(shù)檢測小鼠小腸黏膜固有層中CD_3~+、CD_4~+和CD_8~+T的細胞百分比,采用免疫熒光法檢測各組小鼠小腸黏膜中穿孔素、顆粒酶陽性表達細胞數(shù)。結(jié)果:與正常對照組比較,其他3組小鼠小腸黏膜固有層中CD_4~+T細胞比例均有不同程度下降,CD_8~+T細胞比例均有不同程度上升,有統(tǒng)計學意義(P0.01);正常感染白念組和脾虛感染白念組小鼠小腸黏膜固有層中穿孔素陽性表達細胞均明顯增多(P0.05或P0.01),而顆粒酶陽性表達細胞僅脾虛感染白念組明顯增多(P0.01)。其中,脾虛感染白念組最顯著,與其他3組均具有統(tǒng)計學意義(P0.05或P0.01)。結(jié)論:小腸黏膜固有層中穿孔素和顆粒酶的高表達可能是造成小腸局部損傷的免疫機制之一,脾虛小鼠腸道局部免疫功能下降,感染發(fā)生后可加重腸道局部損傷程度。
[Abstract]:Objective: to observe the effect of Candida albicans infection on T cell subsets, perforin and granzyme in lamina propria of spleen deficiency mice. To explore the partial immune mechanism of Candida albicans infection in spleen deficiency. Methods: the spleen deficiency model of mice was established by the compound factors of diet loss and overwork. The mice were infected with Candida albicans on the 14th day after intestinal infection. Flow cytometry was used to detect the percentage of CD3 ~ + CD4 ~ and CD8 ~ T in the lamina propria of small intestine of mice, and immunofluorescence assay was used to detect the number of perforin and granzyme positive cells in the intestinal mucosa of mice. The percentage of CD4 ~ T cells in the lamina propria of the other three groups decreased in varying degrees, and the percentage of CD8 ~ T cells increased in different degrees. The positive expression of perforin in the lamina propria of small intestine of normal infection group and spleen deficiency group were significantly increased (P 0.05 or P 0.01), while the expression of granzyme positive cells was significantly increased only in the group of spleen deficiency infection (P 0.01), and the expression of perforin in the lamina propria of small intestine of mice infected with spleen deficiency was significantly higher than that in group B (P 0.01). Conclusion: the high expression of perforin and granzyme in the lamina propria of small intestine may be one of the immune mechanisms of local injury of small intestine. The local immune function of spleen deficiency mice decreased, and the degree of local intestinal injury was aggravated after infection.
