【摘要】：目的探討白色脂肪組織(WAT)和棕色脂肪組織(BAT)來源的間充質(zhì)干細(xì)胞成脂分化特性的差異。方法雄性SD大鼠15只,3只用于細(xì)胞分離培養(yǎng),12只用于細(xì)胞移植。體外分離培養(yǎng)WAT和BAT兩種來源的脂肪干細(xì)胞,用油紅O染色檢測兩種干細(xì)胞的成脂分化率,用免疫熒光技術(shù)檢測成脂誘導(dǎo)、分化后的細(xì)胞是棕色脂肪細(xì)胞還是白色脂肪細(xì)胞。4’6-二脒基-2-苯基吲哚(DAPI)標(biāo)記兩種來源的干細(xì)胞后移植至腹股溝區(qū),分別在第1、2、3周取材,免疫熒光技術(shù)檢測植入細(xì)胞的分化趨向。結(jié)果 WAT來源的干細(xì)胞增殖速度明顯快于BAT來源的干細(xì)胞,前者成脂分化率為0.205±0.069,后者為0.165±0.053,兩種干細(xì)胞的成脂誘導(dǎo)率差異無統(tǒng)計(jì)學(xué)意義(P0.05)。兩種干細(xì)胞分別植入體內(nèi)后,在第1、2、3周發(fā)現(xiàn),兩種干細(xì)胞均表達(dá)棕色脂肪特異性蛋白解耦聯(lián)蛋白1(UCP1)。結(jié)論 WAT和BAT來源的干細(xì)胞在體外及體內(nèi)誘導(dǎo)成脂后均分化為棕色脂肪細(xì)胞。
[Abstract]:Objective to investigate the difference of adipogenic differentiation between white adipose tissue (WATT) and brown adipose tissue (BAT). Methods Fifteen male SD rats were used in cell isolation culture and 12 were used for cell transplantation. To culture adipose stem cells from both WAT and BAT, Oil red O staining was used to detect the adipogenic differentiation rate of the two stem cells, and immunofluorescence technique was used to detect the adipogenic induction. The differentiated cells were brown adipocytes or white adipocytes. Results the proliferation rate of stem cells derived from WAT was significantly faster than that of stem cells derived from BAT. The rate of adipogenic differentiation was 0.205 鹵0.069 in the former and 0.165 鹵0.053 in the latter. There was no significant difference in adipogenic induction rate between the two kinds of stem cells. Both stem cells expressed brown adipose-specific protein uncoupling protein 1 (UCP1). Conclusion Stem cells derived from WAT and BAT are evenly divided into brown adipocytes after fat-forming in vitro and in vivo.
