2型糖尿病模型大鼠肝脂代謝相關(guān)基因的篩選及生物信息學(xué)分析(英文)

【摘要】:肝的脂肪代謝異常和胰島素抵抗(insulin resistance,IR)對(duì)促進(jìn)2型糖尿病(type 2 diabetes mellitus,T2DM)的發(fā)生與發(fā)展具有顯著影響。但此過(guò)程復(fù)雜,參與調(diào)控基因目前尚未完全清楚。有研究表明,脂肪酸分解、氨基酸代謝、肝糖原合成等生物過(guò)程對(duì)糖尿病的形成具有促進(jìn)作用。為了闡明這一調(diào)控機(jī)制,本文通過(guò)基因芯片技術(shù)研究GK(Goto-Kakizaki)大鼠和WKY(Wistar-Kyoto)大鼠肝差異基因?qū)Ω蔚闹敬x和胰島素抵抗的影響,探討可引起2型糖尿病發(fā)病的分子機(jī)制。從基因表達(dá)數(shù)據(jù)庫(kù)(GEO)獲取GSE13271基因表達(dá)譜,并對(duì)原始數(shù)據(jù)進(jìn)行標(biāo)準(zhǔn)化處理。通過(guò)GO(Gene Ontology)、KEGG(Kyoto Encyclopedia of Genes and Genomes enrichment)、String和Cytoscape軟件對(duì)差異表達(dá)基因進(jìn)行功能分析。結(jié)果從GK和WKY大鼠中分別獲得179和278個(gè)差異基因,同時(shí)從排名前10的路徑中篩選出21個(gè)差異基因(Aldh1a1,Cyp2c22,Fabp2,Fabp7,Cyp4a3,Acot1,Acot2,Hsd17b2,Ech1,Hmgcl,Bdh1,Crot,Pex11a,Cpt1a,Hadhb,Gda,Elovl2,Prodh,Agpat3,Sardh,Pigu),將這些基因與前10個(gè)的GO term取交集。最終得到10個(gè)顯著差異基因(Aldh1a1,Fabp2,Acot1,Acot2,Ech1,Hmgcl,Bdh1,Crot,Cpt1a,Hadhb),功能分析結(jié)果顯示,肝組織相關(guān)基因通過(guò)一系列生物過(guò)程對(duì)肝的脂肪代謝和胰島素抵抗產(chǎn)生調(diào)節(jié)作用,從而也為臨床糖尿病的治療以及新作用靶點(diǎn)的發(fā)現(xiàn)提供更多參考依據(jù)。
[Abstract]:Abnormal lipid metabolism in liver and insulin resistance IRR have a significant effect on the development and development of type 2 diabetes mellitus type 2 diabetes mellitusus T2DM.However, the process is complex and the genes involved in regulation are not completely clear. Some studies have shown that fatty acid catabolism. Amino acid metabolism, liver glycogen synthesis and other biological processes promote the formation of diabetes mellitus. In this paper, the effects of liver differential genes on fat metabolism and insulin resistance in the liver of GKG Goto-Kakizaki and WKYY Wistar Kyoto rats were studied by gene chip technique, and the molecular mechanism of the pathogenesis of type 2 diabetes was explored. The GSE13271 gene expression profile was obtained from gene expression database. The differential expression genes were analyzed by GO(Gene Ontology Encyclopedia of Genes and Genomes chromosome string and Cytoscape software. Results 179 and 278 differentially expressed genes were obtained from GK and WKY rats, respectively. At the same time, 21 differentially expressed genes, Aldh1a1, Cyp2c22, Fabp2Cyp4a3, Acot1, Acot2Acot2Acot2Acot2Acot2Acot2Acot1, HmgclclBdh1, CrotPex11aCpt1a, HadhbGlovl2Prodhlprodh2Prodh2Sardh term, were screened from the top 10 genes. Finally, 10 distinct genes, Aldh1a, Acot1a, Acot2Acot2Ech1Ech1hmchlhlhchlt1, BhhltCp1t1a, have been obtained. Liver tissue related genes regulate liver fat metabolism and insulin resistance through a series of biological processes, thus providing more references for the treatment of diabetes mellitus and the discovery of new targets.
