流體剪切力對(duì)成骨樣細(xì)胞BMP2和Smad4表達(dá)的實(shí)驗(yàn)研究

【摘要】:目的:探討流體剪切力(FSS)對(duì)骨形態(tài)發(fā)生蛋白2(BMP2)通路中BMP2和Smad4表達(dá)的影響。方法:通過(guò)流體剪切力加載裝置對(duì)體外培養(yǎng)成骨樣細(xì)胞MG63施加12dyn/cm2的流體剪切力Omin,15min,30min,60min和120 min,應(yīng)用免疫熒光雙標(biāo)記法檢測(cè)不同時(shí)間流體剪切力作用于MG63細(xì)胞中BMP2及Smad4的表達(dá)。采用SPSS13.0軟件包對(duì)數(shù)據(jù)進(jìn)行單因素方差分析。結(jié)果:在12dyn/cm2的流體剪切力作用下,MG63成骨樣細(xì)胞中BMP2及Smad4的表達(dá)明顯增多(F=13.532,P=0.000; F=9.058, P=0.001),60min達(dá)高峰;所有組間BMP2表達(dá)有顯著性差異(P0.05);在Omin,15min,30min和120 min組,Smad4的表達(dá)有顯著性差異(P0.05),在60 min組,Smad4的表達(dá)無(wú)顯著性差異(P0.05)。結(jié)論:BMP2通路是流體剪切力信號(hào)轉(zhuǎn)導(dǎo)的重要通路,適宜的流體剪切力刺激通過(guò)BMP2表達(dá)增強(qiáng)而引起Smad4在胞漿內(nèi)積聚,結(jié)合于Smad復(fù)合物,轉(zhuǎn)移至細(xì)胞核,誘導(dǎo)或抑制靶基因的轉(zhuǎn)錄。
[Abstract]:Objective: to investigate the effect of fluid shear stress (FSS) on the expression of BMP2 and Smad4 in bone morphogenetic protein 2 (BMP2) pathway. Immunofluorescence double labeling method was used to detect the expression of BMP2 and Smad4 in MG63 cells exposed to fluid shear stress at different time. The data were analyzed by single factor ANOVA using SPSS13.0 software package. Results: MG63 was formed by fluid shear stress of 12 dyn / cm ~ 2. The expression of BMP2 and Smad4 in osteoid cells increased significantly (P < 0.05), the expression of BMP2 and Smad4 increased significantly (P < 0.05), and the expression of BMP2 and Smad4 increased significantly (P < 0.05), and the expression of BMP2 and Smad4 increased significantly (P < 0.05). There was significant difference in the expression of BMP2 between all groups (P 0.05), in 30 minutes after 15 mins and 120 min, there was a significant difference in the expression of Smad4, but there was no significant difference in the expression of Smad4 in 60 min group. Conclusion the expression of BMP2 is an important pathway in signal transduction of fluid shear stress, and there is no significant difference in the expression of Smad4 in 60 min group. Appropriate fluid shear stress stimuli induce the accumulation of Smad4 in the cytoplasm through enhanced BMP2 expression, bind to the Smad complex, transfer to the nucleus and induce or inhibit the transcription of the target gene.
