沖擊波對(duì)人骨髓間充質(zhì)干細(xì)胞增殖和成骨分化作用的研究

【摘要】:目的:探討沖擊波對(duì)人骨髓間充質(zhì)干細(xì)胞(MSC)增殖和成骨分化能力的影響。方法:從健康志愿者骨髓中分離培養(yǎng)骨髓間充質(zhì)干細(xì)胞,將其分為3組,空白對(duì)照組細(xì)胞用普通培養(yǎng)液培養(yǎng),成骨誘導(dǎo)組細(xì)胞用經(jīng)典成骨誘導(dǎo)劑培養(yǎng),沖擊波組細(xì)胞用普通培養(yǎng)液培養(yǎng),外加沖擊波刺激。刺激后在鏡下觀察MSC形態(tài);應(yīng)用CCK-8檢測(cè)沖擊波組細(xì)胞和空白對(duì)照組MSC增殖能力的變化;第14天和28天分別用堿性磷酸酶和Von Kossa染色檢測(cè)各組細(xì)胞的成骨能力。結(jié)果:CCK-8檢測(cè)發(fā)現(xiàn),沖擊波與空白對(duì)照組相比,細(xì)胞的增殖活性顯著增強(qiáng);堿性磷酸酶和Von Kossa染色結(jié)果表明,沖擊波組細(xì)胞的成骨分化能力比成骨誘導(dǎo)組要高;空白對(duì)照組細(xì)胞染色呈現(xiàn)弱陽(yáng)性。結(jié)論:沖擊波能促進(jìn)骨髓間充質(zhì)干細(xì)胞的增殖能力和成骨分化能力。
[Abstract]:Objective: to investigate the effect of shock wave on the proliferation and osteogenic differentiation of human bone marrow mesenchymal stem cells (MSCs). Methods: bone marrow mesenchymal stem cells were isolated from healthy volunteers and divided into 3 groups. The cells of blank control group were cultured in ordinary culture medium, the cells of osteoblast induction group were cultured with classical osteogenic inducer, and the cells of shock wave group were cultured in ordinary culture medium. The morphology of MSC was observed under microscope after shock wave stimulation. CCK-8 was used to detect the proliferation ability of MSC in shock wave group and blank control group. Alkaline phosphatase and Von Kossa staining were used to detect the osteogenic ability of the cells in each group on the 14th and 28th days. Results the shock wave was compared with the control group. The proliferative activity of the cells was significantly increased. The results of alkaline phosphatase and Von Kossa staining showed that the osteogenic differentiation ability of shock wave group was higher than that of osteoblast induction group. Conclusion: shock wave can promote the proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells.
