大鼠骨髓間充質(zhì)干細胞聯(lián)合α-干擾素對大鼠肝損傷的治療作用

【摘要】:目的:1建立大鼠肝損傷模型。 2在大鼠肝損傷模型的基礎(chǔ)上初步探討大鼠骨髓間充質(zhì)干細胞聯(lián)合α-干擾素對肝損傷的治療作用。 方法:1用四氯化碳油溶液(50%體積分數(shù))按2ml/kg對SD大鼠行腹腔注射,每周兩次,共6周,建立大鼠肝損傷模型。于6周后隨機挑選6只,處死大鼠,取肝組織行HE染色評價肝損傷程度。 2將肝損傷模型大鼠隨機分為空白對照組(K)、大鼠骨髓間充質(zhì)干細胞治療組(S)、干擾素組(I)、大鼠骨髓間充質(zhì)干細胞+干擾素治療組(C)。分別于治療后3天,6天處死大鼠,采血檢測谷草轉(zhuǎn)氨酶(AST)、谷丙轉(zhuǎn)氨酶(ALT)、總膽紅素(TBIL)、HA透明質(zhì)酸酶、LN層粘連蛋白、PCⅢ型前膠原、Ⅳ型膠原,同時取肝組織行HE染色評價肝損傷嚴重程度。 結(jié)果:1大鼠肝損傷模型60只,死亡6只,存活54只死亡率為10%,存活的54只均出現(xiàn)腹圍增加,隨機選取6只取肝臟HE染色均提示有不同程度肝損傷,成功率為90%,符合本實驗要求。 2大鼠慢性肝損傷后,AST指標顯著高于ALT指標;藥物治療后,除了ALT濃度的干細胞治療組和干擾素治療組無差異外,AST、ALP、TBDL、LN,Ⅳ型膠原的組間對比均有顯著性差異,聯(lián)合治療組濃度低于其余三組,且具有統(tǒng)計學差異。ALT,HA和Ⅲ型前膠原的濃度,聯(lián)合治療組濃度低于其余三組,且具有統(tǒng)計學差異,空白對照組和干細胞組相比有統(tǒng)計學差異,干細胞治療組濃度低于空白對照組,干細胞治療組濃度低于干擾素組,有統(tǒng)計學差異,空白對照組和干擾素組相比無統(tǒng)計學差異。 結(jié)論:1四氯化碳油溶液(50%體積分數(shù))按2ml/kg對大鼠行腹腔注射,每周兩次,共6周,建立肝損傷模型穩(wěn)定可靠,為較理想的研究肝損傷模型。2α-干擾素和大鼠骨髓間充質(zhì)干細胞對四氯化碳誘導的大鼠肝損傷有一定的治療作用,干細胞治療效果優(yōu)于干擾素。3大鼠骨髓間充質(zhì)干細胞聯(lián)合α-干擾素對四氯化碳誘導的肝損傷的治療有協(xié)同效果,聯(lián)合治療效果優(yōu)于單藥治療。
[Abstract]:Objective to establish a rat model of liver injury by 1: 1. 2 on the basis of rat liver injury model, the therapeutic effect of bone marrow mesenchymal stem cells combined with interferon on liver injury was studied. Methods Sprague-Dawley rats were injected intraperitoneally with 50% volume fraction of carbon tetrachloride oil solution twice a week for 6 weeks. Six rats were randomly selected after 6 weeks. Rats were killed and liver tissues were taken to evaluate the degree of liver injury by HE staining. 2Rats with liver injury were randomly divided into control group (control group), rat bone marrow mesenchymal stem cell (BMSCs) treatment group and interferon group (group I). The rat bone marrow mesenchymal stem cell interferon treatment group was killed on 3 and 6 days after treatment, and blood samples were collected to detect aspartate and alanine aminotransferase (alt).
