三步序貫法對哮喘大鼠模型激素撤減中轉(zhuǎn)化生長因子β1及膠原表達(dá)的影響

【摘要】:目的:觀察三步序貫法對哮喘模型大鼠激素撤減過程中轉(zhuǎn)化生長因子-β(T G F-β)及膠原(Collagen)表達(dá)的影響。方法:將150只雄性SD大鼠隨機(jī)分為正常對照組、氣道重塑組、地塞米松組、普米克令舒組及三步序貫組。模型建立方法:以卵蛋白100mg加氫氧化鋁100mg致敏,1%卵蛋白激發(fā),自激發(fā)之日開始,于激發(fā)前30min給予地塞米松0.5mg/kg腹腔注射,連續(xù)2周,從第3周開始,按每周0.1mg/kg速度遞減地塞米松用量,至第7周地塞米松全部撤除。三步序貫組在地塞米松干預(yù)基礎(chǔ)上,于實(shí)驗(yàn)第15-28天(地塞米松撤前)給予第1步方顆粒灌胃,第29-63天(地塞米松撤中)給予第2步方顆粒灌胃,第64-77天(地塞米松撤后)給予第3步方顆粒灌胃,每日1次。各組于實(shí)驗(yàn)流程第28、63、77天末次激發(fā)2h后取左肺組織行免疫組化檢測,比較各組TGF-β1、CollagenⅠ、CollagenⅢ蛋白表達(dá)變化。結(jié)果:在撤減過程中,與正常對照組比較,氣道重塑組和地塞米松組CollagenⅠ蛋白表達(dá)升高(P0.05),TGF-β1、CollagenⅢ均顯著升高(P0.01);與地塞米松組比較,三步序貫組TGF-β1、CollagenⅠ顯著降低(P0.01,P0.05),CollagenⅢ在撤減中后期降低(P0.05)。結(jié)論:三步序貫法對哮喘模型大鼠撤減激素過程中氣道重塑的阻抑作用可能是通過降低TGF-β1,調(diào)控其介導(dǎo)的信號通路,進(jìn)而減少CollagenⅠ和CollagenⅢ沉積來實(shí)現(xiàn)的。
[Abstract]:Objective: to observe the effect of three-step sequential method on transforming growth factor- 尾 (TGF- 尾) and collagen collagen during hormone withdrawal in asthmatic rats. Methods: 150 male SD rats were randomly divided into normal control group. Airway remodeling group, dexamethasone group, Pulmicort group and three-step sequential group. Methods: ovalbumin 100 mg + aluminum hydroxide 100 mg sensitized with 1% ovalbumin was induced from the day of stimulation. Dexamethasone 0.5 mg / kg was injected intraperitoneally 30 minutes before challenge for 2 weeks. From the third week, the dosage of dexamethasone was decreased at the rate of 0.1 mg / kg per week. By week 7, dexamethasone was completely removed. On the basis of dexamethasone intervention, the three-step sequential group was given intragastric infusion of Dexamethasone granule on the 15-28 days (before dexamethasone withdrawal).
