Hepcidin、鐵和炎癥相互關(guān)系的初步研究

【摘要】:Hepcidin是一種抗菌肽,也是重要的鐵調(diào)素分子。在高等脊椎動(dòng)物中研究發(fā)現(xiàn)它不僅能直接抑制細(xì)菌生長(zhǎng),還可以調(diào)控機(jī)體的鐵代謝平衡。它既在機(jī)體天然免疫中發(fā)揮作用,又在機(jī)體內(nèi)環(huán)境穩(wěn)定中擔(dān)當(dāng)角色,是一個(gè)重要的界面分子。Hepcidin的表達(dá)受多種因素影響,炎癥反應(yīng)就是其中之一。本論文我們以CpG刺激斑馬魚(yú)來(lái)構(gòu)建炎癥反應(yīng)模型,研究Hepcidin、鐵和炎癥之間的相互關(guān)系。研究過(guò)程中我們用CpG-ODN、含CpG重復(fù)基元的pYES質(zhì)粒和嗜水氣單胞菌(Aeromonas hydrophila)基因組DNA刺激斑馬魚(yú),利用RT-PCR和熒光定量PCR檢測(cè)刺激前后Hepcidin表達(dá)的變化,統(tǒng)計(jì)斑馬魚(yú)在含CpG質(zhì)粒預(yù)刺激再感染嗜水氣單胞菌后的死亡率,并進(jìn)一步檢測(cè)斑馬魚(yú)血清中Hepcidin及鐵含量的變化,同時(shí)初步探索了這兩者以及補(bǔ)體的變化對(duì)機(jī)體抗細(xì)菌感染能力的影響。研究結(jié)果顯示CpG刺激后斑馬魚(yú)肝臟Hepcidin mRNA表達(dá)上調(diào);嗜水氣單胞菌感染后的存活率顯著提高;血清中Hepcidin濃度提高,但血清中的鐵含量下降;血清抑制細(xì)菌生長(zhǎng)的能力和Hepcidin、鐵還有補(bǔ)體有關(guān)。 研究表明炎癥發(fā)生時(shí)Hepcidin表達(dá)上調(diào),而血清鐵含量下降,進(jìn)而抑制細(xì)菌生長(zhǎng),從而抑制體內(nèi)的炎癥反應(yīng),這是宿主體內(nèi)一種抑制細(xì)菌感染的新機(jī)制,研究結(jié)果為深入探討Hepcidin在機(jī)體中的抑菌機(jī)制打下基礎(chǔ)。
[Abstract]:Hepcidin is an antimicrobial peptide and an important iron modulin molecule. Studies in higher vertebrates have found that it not only directly inhibits bacterial growth. It can also regulate the balance of iron metabolism in the body. It not only plays a role in the innate immunity of the body, but also plays a role in the stability of the body environment. Inflammatory response is one of the most important interfacial molecules, and inflammatory response is one of them. In this paper, we use CpG to stimulate zebrafish to construct inflammatory response model. To study the relationship between hepcidin, iron and inflammation. We used CpG-ODN in the course of the study. Zebrafish were stimulated by pYES plasmid containing CpG repeats and Aeromonas hydrophila genomic DNA. The changes of Hepcidin expression before and after stimulation were detected by RT-PCR and fluorescence quantitative PCR, and the mortality of zebrafish after pre-stimulation with CpG plasmid and re-infection with Aeromonas hydrophila was analyzed. The changes of Hepcidin and iron in zebrafish serum were further determined. At the same time, the effects of these two and the changes of complement on the ability of anti-bacterial infection were preliminarily explored. The results showed that the expression of Hepcidin mRNA in the liver of zebrafish was up-regulated after CpG stimulation. The survival rate of Aeromonas hydrophila was significantly increased after infection. The concentration of Hepcidin in serum increased, but the content of iron in serum decreased. The ability of serum to inhibit bacterial growth is related to hepcidin, iron and complement. Studies have shown that Hepcidin expression is up-regulated and serum iron content is decreased during inflammation, which inhibits bacterial growth and thus inhibits the inflammatory response in vivo. This is a new mechanism to inhibit bacterial infection in the host. The results lay a foundation for the further study of the bacteriostatic mechanism of Hepcidin in the organism.
