【摘要】：目的觀察基底膜蛋白多糖對體外培養(yǎng)的胎鼠顱骨成骨樣細胞合成分泌轉(zhuǎn)化生長因子β1的影響。 方法分離并培養(yǎng)SD胎鼠顱蓋骨成骨樣細胞,采用堿性磷酸酶染色法及茜素紅染色法鑒定培養(yǎng)細胞性質(zhì),于第3,5,7,9,11,13天時收集上清液,隨即分對照組、基底膜蛋白多糖阻斷組,應(yīng)用免疫組織化學(xué)染色及酶聯(lián)免疫吸附試驗檢測基底膜蛋白多糖對成骨細胞分泌轉(zhuǎn)化生長因β1的表達及濃度變化。 結(jié)果對照組基底膜蛋白多糖免疫組化呈強陽性表達。與對照組比較,基底膜蛋白多糖阻斷組的轉(zhuǎn)化生長因子β1濃度降低(P0.05),差異有顯著性意義。兩組生長因子的表達量及濃度均于第7天時最高,而后逐漸下降。 結(jié)論基底膜蛋白多糖抗體消除其促進成骨細胞分泌合成轉(zhuǎn)化生長因子β1的作用,使轉(zhuǎn)化生長因子β1濃度降低。
[Abstract]:Objective To observe the effect of basilar membrane proteoglycan on the synthesis and secretion of transforming growth factor beta 1 in cultured fetal rat calvarial osteoblast like cells.
Methods SD were isolated and cultured fetal rat calvarial osteoblasts, alkaline phosphatase staining and alizarin red staining was used to identify the cultured cells in nature, in the 3,5,7,9,11,13 days when the supernatant was collected, and then divided into control group, perlecan block group, immunohistochemical staining and enzyme linked immunosorbent assay of basement membrane proteins the test of polysaccharide on osteoblast secretion of transforming growth factor beta 1 expression and concentration.
Results in the control group, basement membrane proteoglycan immunohistochemistry showed strong positive expression. Compared with the control group, basement membrane proteoglycan block by transforming growth factor beta 1 concentration decreased (P0.05), there was a significant difference between the two groups. The expression amount and concentration of growth factors on the seventh day of the highest, then gradually decreased.
Conclusion basilar membrane proteoglycan antibody eliminates the effect on the secretion of transforming growth factor beta 1 from osteoblasts and reduces the concentration of TGF - beta 1.
