【摘要】：目的:觀察筋骨痛消丸干預(yù)后膝OA模型兔膝關(guān)節(jié)液中IL-1、TNF-α含量的變化,探討筋骨痛消丸治療膝骨性關(guān)節(jié)炎的作用機(jī)制。方法:將42只新西蘭兔隨機(jī)抽出11只納入空白組,余納入模型組,模型組用管狀石膏伸直位固定右后膝5周制作膝骨性關(guān)節(jié)炎模型,完成造模后,隨機(jī)抽取空白組和模型組各1只兔子處死以驗(yàn)證模型成功后,拆除造模組石膏。將沒(méi)有造模的剩余10只兔子分為A組(空白組),造模成功的剩余30只兔子隨機(jī)分為B組(模型組)、C組(筋骨痛消丸組)、D組(鹽酸氨基葡萄糖組)。C組予以與60Kg體重人等效劑量的筋骨痛消丸懸濁液10mL、D組予以與60Kg體重人等效劑量的鹽酸氨基葡萄糖混懸液10mL灌胃,A、B組予以10mL蒸餾水灌胃,灌胃處理3周后處死,行各組肉眼觀察膝關(guān)節(jié)大體形態(tài)以及取股骨髁軟骨行光鏡觀察,ELISA法測(cè)定膝關(guān)節(jié)液中IL-1以及TNF-α含量。結(jié)果:①造模5周后,造模組膝關(guān)節(jié)出現(xiàn)明顯的退行性改變。②治療后肉眼觀察B、C、D組較A組相比均有退變,C、D組的退變程度較B組輕。光鏡下B、C、D組均可見(jiàn)軟骨退變的病理變化。③治療后:B、C、D組與A組相比較關(guān)節(jié)液中IL-1和TNF-α含量明顯升高(P0.05).C、D組關(guān)節(jié)液中IL-1和TNF-α含量與B組相比有明顯減少(P0.05),治療后C組IL-1、TNF-α含量與D組比較無(wú)顯著差異(P0.05)。結(jié)論:筋骨痛消丸對(duì)兔膝骨性關(guān)節(jié)炎有防治作用,其機(jī)理可能是通過(guò)降低關(guān)節(jié)液中細(xì)胞炎性因子IL-1、TNF-α含量從而抑制軟骨的破壞,延緩骨性關(guān)節(jié)炎的進(jìn)展。
[Abstract]:Objective: the rabbit model of knee OA in knee joint fluid observation of Jingu Tongxiao pills after the intervention of IL-1, changes of TNF- alpha content, to explore the mechanism of action of Jingu Tongxiao pills in the treatment of knee osteoarthritis. Methods: 42 New Zealand rabbits were randomly selected 11 rats into control group, more than in model group, model group with tubular gypsum right knee unbend fixed after 5 weeks for knee osteoarthritis model, complete model, randomly selected control group and model group with 1 rats in each group rabbits were sacrificed to verify the model after the success of the model.
