四種重要醫(yī)學(xué)媒介蚊蟲離子受體基因IR8a和IR25a的特征及分類地位

【摘要】:【目的】鑒定中華按蚊Anopheles sinensis、岡比亞按蚊Anopheles gambiae、埃及伊蚊Aedes aegypti和致倦庫蚊Culex quinquefasciatus 4種重要醫(yī)學(xué)媒介蚊蟲的離子受體基因IR8a與IR25a以及代表性的iGluRs基因和IRs基因,研究這些基因的特征及其系統(tǒng)發(fā)育關(guān)系,確定IR8a與IR25a基因在離子谷氨酸受體基因中的分類地位。【方法】以黑腹果蠅Drosophila melanogaster的iGluRs和IRs基因序列作為詢問序列,用BLASTP和BLASTN方法分別搜索中華按蚊、岡比亞按蚊、埃及伊蚊和致倦庫蚊的基因組數(shù)據(jù)庫,以鑒定這4個(gè)種內(nèi)的iGluRs和IRs基因序列;用生物信息學(xué)方法比較分析鑒定獲得的IR8a與IR25a及代表性的iGluRs和IRs基因氨基酸序列的特征;用最大似然法、貝葉斯法和最大簡(jiǎn)約法構(gòu)建并討論這些基因氨基酸序列的系統(tǒng)發(fā)育關(guān)系;用PAML軟件計(jì)算這些基因的dN/dS(ω)值并分析其選擇壓力!窘Y(jié)果】通過對(duì)中華按蚊、岡比亞按蚊、埃及伊蚊和致倦庫蚊中離子谷氨酸受體基因進(jìn)行生物信息學(xué)鑒定,獲得了所有IR8a和IR25a基因,24個(gè)代表性的iGluRs基因和30個(gè)代表性的IRs基因的序列。特征比較和系統(tǒng)發(fā)育關(guān)系分析結(jié)果表明,在4種蚊蟲和黑腹果蠅中IR8a與IR25a基因的氨基酸序列長(zhǎng)度與iGluRs的更接近,其平均值遠(yuǎn)大于其他IRs的;IR25a和IR8a有與傳統(tǒng)iGluRs相同的氨基酸末端區(qū)域(amino-terminal domain,ATD),雖然IR8a的序列不太保守,而IRs則沒有ATD;IR8a與IR25a序列有與傳統(tǒng)iGluRs一樣保守的配體結(jié)合區(qū)S1和S2,而IRs沒有。所有這些基因的ω值遠(yuǎn)小于1,意味著這些基因在進(jìn)化過程中都經(jīng)歷了純化選擇,而IR8a與IR25a的ω值更接近iGluRs。此外,IR8a與IR25a還有與non-NMDA相似的特異性位點(diǎn),而這些位點(diǎn)在IRs不存在。再者,IR8a和IR25a與non-NMDA聚在一枝,形成一個(gè)姐妹群,群內(nèi)序列間的遺傳距離小于本研究中任何其他序列間的距離。根據(jù)這些研究結(jié)果,我們將IR8a與IR25a分類進(jìn)入傳統(tǒng)iGluRs中的non-NMDA類型,并命名為Putative受體!居懻摗勘狙芯繕(gòu)建了蚊蟲iGluRs和IRs基因的信息框架,確定了IR8a與IR25a基因的分類地位,對(duì)進(jìn)一步的功能研究具有重要意義。
[Abstract]:[Objective] identification of Anopheles sinensis Anopheles sinensis, Gambia Anopheles gambiae Aedes mosquito Anopheles, Culex Culex aegypti and quinquefasciatus 4 kinds of important medical vector mosquito ion receptor gene IR8a and IR25a and the representative of the iGluRs gene and IRs gene, phylogenetic relationships of the characteristics and system of these genes, determine the taxonomic status of IR8a and IR25a gene the ionotropic glutamate receptors genes. [method] with iGluRs and IRs gene sequences of Drosophila melanogaster Drosophila melanogaster as query sequences, the search of Anopheles sinensis, respectively using BLASTP and BLASTN methods in Gambia Anopheles, Culex quinquefasciatus and Aedes aegypti genome database, with iGluRs and IRs gene sequences of 4 species in this identification; by comparative analysis of biological information science methods identified with IR25a and IR8a on behalf of iGluRs and the amino acid sequence of IRs gene Characteristics; maximum likelihood, Bayesian and maximum parsimony method is constructed and discussed the relationship between the amino acid sequences of these genes system development; calculation of these genes using dN/dS PAML software (omega) value and analyze the selection pressure. [results] by Anopheles sinensis, Gambia Anopheles, Aedes aegypti and induced by glutamate receptor ion in Culex quinquefasciatus gene bioinformatics identification, access to all the IR8a and IR25a gene, iGluRs gene sequences of 24 representative and 30 representative IRs genes. Comparison of the characteristics and relationship between the phylogenetic analysis results showed that the amino acid sequence of iGluRs and IR8a and IR25a gene in 4 species of mosquitoes and insects in Drosophila melanogaster. Closer, the average value is far greater than other IRs; IR25a and IR8a have the same amino acids with the traditional iGluRs (amino-terminal domain, ATD terminal region), although the IR8a sequence is not too conservative, but not IRs, ATD; I R8a and IR25a have the same sequence with traditional iGluRs ligand binding region conserved S1 and S2, but not IRs. All of these gene values far less than 1, means that these genes in the evolutionary process have experienced negative selection, and the value of IR8a and omega IR25a is close to iGluRs. IR8a and IR25a and in addition, similar to non-NMDA the specific sites, and these sites do not exist in IRs. Furthermore, IR8a and IR25a and non-NMDA were clustered in the same branch, formed a sister group, the genetic distance between sequences within the group is less than the distance between any other sequences in this study. According to these results, we will IR8a and IR25a into the traditional iGluRs classification the type of non-NMDA, and named as Putative receptor. [Conclusion] this study constructs the information framework mosquito iGluRs and IRs genes, determine the taxonomic status of IR8a and IR25a gene, has important significance for the further research on the function.
