【摘要】:目的觀察小鼠克羅恩病模型外周血單核細(xì)胞及結(jié)腸組織中白細(xì)胞介素(IL)17、叉頭狀/翅狀螺旋蛋白3(Foxp3)的表達(dá),探討其在2,4,6-三硝基苯磺酸(TNBS)誘導(dǎo)的克羅恩病小鼠急性炎癥中的作用。方法共40只6~8周齡雌性Bal/c小鼠,采用隨機(jī)數(shù)字表法隨機(jī)分為兩組,20只以TNBS/乙醇溶液灌腸誘導(dǎo)克羅恩病模型(實(shí)驗(yàn)組),另外20只小鼠以同等濃度的乙醇氯化鈉灌腸(正常對(duì)照組)。干預(yù)1周后行疾病活動(dòng)指數(shù)(DAI),結(jié)腸病理組織學(xué)評(píng)分(TDI)和結(jié)腸大體形態(tài)損傷指數(shù)(CMDI)評(píng)價(jià),以實(shí)時(shí)熒光定量反轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)檢測(cè)小鼠外周血單核細(xì)胞IL-17mRNA、Foxp3 mRNA的表達(dá),采用免疫印跡法檢測(cè)小鼠結(jié)腸組織IL-17蛋白、Foxp3蛋白的表達(dá)。結(jié)果與正常對(duì)照組相比,實(shí)驗(yàn)組的DAI、TDI、CMDI顯著升高[DAI(5.41±0.48)比(0.85±0.07),TDI(5.63±0.74)比(0.74±0.08),CMDI(6.88±0.84)比(0.91±0.06),P0.01],外周血單核細(xì)胞的IL-17 mRNA顯著升高[(1.118±0.145)比(1.000±0.000),P0.01],與其DAI、TDI及CMDI呈正相關(guān)(r=0.875,0.935,0.825,P0.01),Foxp3 mRNA顯著降低[(0.797±0.271)比(1.000±0.000),P0.01],與其DAI、TDI及CMDI呈負(fù)相關(guān)(r=-0.885,-0.734,r-0.812,P0.01),結(jié)腸組織IL-17蛋白顯著上升[(0.631±0.331)比(0.272±0.112),P0.01],與其DAI、TDI及CMDI呈正相關(guān)(r=0.765,0.778,0.734,P0.01),Foxp3蛋白顯著降低[(0.274±0.112)比(0.592±0212),P0.01],與DAI、TDI及CMDI呈負(fù)相關(guān)(r=-0.889,-0.809,-0.851,P0.01)。結(jié)論在TNBS誘導(dǎo)的結(jié)腸炎中,Th17/Treg軸的免疫偏倚是導(dǎo)致克羅恩病發(fā)生和發(fā)展的重要因素。
[Abstract]:Objective To observe the effect of Crohn's disease model mice peripheral blood mononuclear cells and colon tissue of interleukin 17 (IL), forkhead / winged helix protein 3 (Foxp3) expression on 2,4,6- three trinitrobenzene sulfonic acid (TNBS) in mice induced by acute inflammation in Crohn's disease in rats. Methods a total of 40 only 6~8 week old female Bal/c mice were randomly divided into two groups, 20 rats in TNBS/ ethanol solution enema induced model of Crohn's disease (experimental group), another 20 mice with the same concentration of sodium chloride and ethanol enema (normal control group). The intervention was performed after 1 weeks of disease activity index (DAI). Colon histopathology score (TDI) and colon gross morphological damage index (CMDI) evaluated by real-time fluorescent quantitative reverse transcription polymerase chain reaction (RT-PCR) IL-17mRNA in monocytes in peripheral blood to detect expression of Foxp3, mRNA, the immune blotting detection of colon tissue of mice IL-17 铔嬬櫧,Foxp3铔嬬櫧鐨勮〃杈,
