富血小板血漿(PRP)對(duì)大鼠游離脂肪移植物存活作用的實(shí)驗(yàn)研究

【摘要】:目的建立wistar大鼠游離脂肪移植的實(shí)驗(yàn)動(dòng)物模型,觀(guān)察富血小板血漿(PRP)對(duì)游離脂肪移植物存活的作用。 方法實(shí)驗(yàn)采用自身對(duì)照,獲取近交系wistar大鼠脂肪組織和PRP,于24只wistar大鼠背部分別注射以下三組脂肪移植物:A空白對(duì)照組(n=24),0.5ml脂肪組織+0.12ml生理鹽水;B實(shí)驗(yàn)組1(PRP未激活組,n=24),0.5ml脂肪組織+0.1ml PRP+0.02ml生理鹽水;C實(shí)驗(yàn)組2(PRP激活組,n=24),0.5ml脂肪組織+0.1ml PRP+0.02ml0.9%氯化鈣。移植術(shù)后1月、2月、3月各處死8只大鼠,分別獲取1月、2月和3月三組移植物標(biāo)本,進(jìn)行形態(tài)學(xué)和組織學(xué)觀(guān)察,標(biāo)本行HE染色,于光鏡下觀(guān)察脂肪組織存活情況,包括脂肪組織完整性、囊腔和空泡的形成、纖維化程度、炎癥反應(yīng)和血管密度,分別從以上方面進(jìn)行病理學(xué)評(píng)分,采用配對(duì)t檢驗(yàn)(SPSS13.0)對(duì)數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析。 結(jié)果PRP中血小板數(shù)(4871±635.5)明顯高于全血中血小板數(shù)(617.6±61.30)(P<0.0001);形態(tài)學(xué)觀(guān)察結(jié)果:移植后脂肪組織體積逐漸變小,質(zhì)地變硬,周?chē)纬梢粚影,表面可?jiàn)血管網(wǎng)形成,術(shù)后1月,部分脂肪移植物已有明顯吸收,,但無(wú)明顯壞死、液化、膿腫形成等并發(fā)癥發(fā)生,術(shù)后2月和3月時(shí),已無(wú)法觸及明顯移植物,甚至完全消失不見(jiàn);術(shù)后1月移植物體積比較,三組間均無(wú)明顯差異(P>0.05),而PRP激活組移植物直徑明顯大于空白組(P<0.05);組織學(xué)觀(guān)察結(jié)果:術(shù)后1月,PRP激活組的血管密度明顯高于空白對(duì)照組(P<0.05);術(shù)后3月,PRP激活組的炎癥反應(yīng)明顯小于PRP未激活組和空白組(P<0.05);而其他病理觀(guān)察指標(biāo):脂肪組織完整性、纖維化程度及囊腔空泡形成,在各時(shí)間段三組間均無(wú)統(tǒng)計(jì)學(xué)差異(P>0.05)。 結(jié)論在wistar大鼠模型上PRP對(duì)游離脂肪移植物的存活無(wú)明顯促進(jìn)作用。
[Abstract]:Objective to establish an experimental animal model of free fat transplantation in wistar rats and to observe the effect of platelet-rich plasma wistar on the survival of free fat grafts. Methods the adipose tissue and PRP of inbred wistar rats were obtained by self-control. The following three groups of fat grafts were injected into the back of 24 wistar rats. In the control group, 0. 12 ml of normal saline was injected into 0. 12 ml of adipose tissue. In group B, 0. 1 ml PRP 0. 02 ml normal saline was found in 0. 5 ml of adipose tissue. In group C, 0.1 ml PRP 0.02 ml 0.9% calcium chloride was found in 0. 5 ml adipose tissue in group 2 of PRP activation. January, February after transplantation. On March, 8 rats were killed and grafts were collected from January, January and March 3rd groups respectively. Morphological and histological observation was performed. The specimens were stained with HE, and the survival of adipose tissue was observed under light microscope. Including adipose tissue integrity, cystic and vacuolar formation, degree of fibrosis, inflammatory response and vascular density, pathological scores were performed from the above aspects. The data were statistically analyzed by paired t test (SPSS 13.0). Results the platelet count in PRP (4871 鹵635.5) was significantly higher than that in whole blood (617.6 鹵61.30) (P < 0.0001). Morphological observation: after transplantation, the volume of adipose tissue gradually became smaller, the texture became hard, a coating was formed around it, and the vascular network was formed on the surface. On January, some fat grafts had been absorbed obviously. However, there was no obvious necrosis, liquefaction, abscess and other complications. On February and March, the grafts could not be reached or even disappeared completely. On January, there was no significant difference in graft volume among the three groups (P > 0.05), but the diameter of graft in PRP activated group was significantly larger than that in blank group (P < 0.05). Results of histological observation: on January, the vascular density in PRP activated group was significantly higher than that in blank control group (P < 0.05). On March, the inflammatory response in PRP-activated group was significantly lower than that in non-activated PRP group and blank group (P < 0.05). Other pathological indicators: adipose tissue integrity, degree of fibrosis and cavitation formation were not significantly different among the three groups (P > 0.05). Conclusion PRP has no effect on the survival of free fat grafts in wistar rats.
