【摘要】:目的建立巴貝西蟲感染的黑線倉鼠模型,明確感染后黑線倉鼠生物學特性的變化規(guī)律,為巴貝西蟲病的檢測與防治提供基礎數(shù)據(jù)資料。方法腹腔注射含巴貝西蟲的血液感染黑線倉鼠,感染的第0、2、4、6、8、10、12、14、16、23、30、37天每次取5只動物采集抗凝血和全血,制備血涂片,通過吉姆薩染色檢測蟲體繁殖情況;分離血液總DNA,用REAL-TIME PCR檢測巴貝西蟲的在宿主體內(nèi)的繁殖規(guī)律;用全自動生理生化檢測儀測定血液生理生化指標;處死動物后分別采集心、肝、脾、肺、腎等器官,稱重測定臟器系數(shù);用ELISA法檢測感染動物血清中IL-2濃度。結果感染后第4天黑線倉鼠體內(nèi)的巴貝西蟲數(shù)量最多,之后整體呈下降趨勢,在第12天有一個短暫升高。感染動物的臟器系數(shù)變化最大的屬于肝臟和脾臟,心、肺和腎臟系數(shù)在整個感染期內(nèi)稍有波,均在正常值范圍內(nèi)。感染動物的血細胞均有波動,在第10、23天兩次達峰值,其中白細胞變化最為劇烈;檢測到變化的血液生化指標在第12天達峰值。感染動物血清中的IL-2在第10天達峰值,之后連續(xù)下降。結論感染巴貝西蟲的黑線倉鼠具有典型的蜱傳寄生蟲病特點,病原侵入一周內(nèi)達繁殖頂峰,且病原可在宿主體內(nèi)長期潛伏。宿主免疫響應在第2周達到峰值,與免疫相關的臟器及血細胞有明顯的應激反應。據(jù)此,可有針對性的開展巴貝西蟲病的診斷與防治。
[Abstract]:Objective to establish a model of Cricetulus BABEI West infection after infection, clear Cricetulus biological characteristics, and provide basic information for the detection and prevention of BABEI West worm disease. Methods of intraperitoneal injection with BABEI West insect blood infection Cricetulus barabansis, each day 0,2,4,6,8,10,12,14,16,23,30,37 infection in 5 animal anti coagulation and blood collection preparation, blood smear, detection of parasite reproduction by Giemsa staining; separation of blood DNA, REAL-TIME PCR detection of BABEI west of worms in the propagation of the host; Determination of physiological and biochemical indexes of blood by automatic physiological and biochemical detection instrument; animal were sacrificed respectively after collecting heart, liver, spleen, lung, kidney and other organs weighing, determination of organ coefficient; ELISA method was used to detect the concentration of IL-2 infection in animal serum. Results fourth days after infection in the number of Cricetulus BABEI West worms, after a whole A downward trend, on the twelfth day there was a transient increase in infected animal organs. The maximum coefficient of variation belongs to the liver and spleen, heart, lung and kidney coefficient in the infection period a little wave, are within the normal range. All blood cell fluctuations of infected animal, in the peak two days 10,23. The most dramatic changes in white blood cells; blood biochemical index changes were detected in twelfth Tianda peak. Infection in animal serum IL-2 in the tenth Tianda peak, after a continuous decline. Conclusion the infection BABEI West insect Cricetulus with typical tick borne parasitic disease characteristics, pathogen invasion and reproduction within a week peak, long-term pathogen lurking in the host. The host immune response reached a peak in second weeks, and immune related organs and blood cell stress reaction obviously. Accordingly, there may be in order to carry out the diagnosis and treatment of echinococcosis of the west of BABEI.
