【摘要】:目的探討構(gòu)建尿酸排泄不良型高尿酸血癥大鼠模型的可靠方法,為尿酸排泄不良型高尿酸血癥發(fā)病機(jī)制的研究及治療方案的優(yōu)化奠定基礎(chǔ)。方法采用氧嗪酸鉀(300 mg/kg)分別與吡嗪酰胺(300 mg/kg)、乙胺丁醇(250 mg/kg)聯(lián)合造模。連續(xù)給藥1周、3周、5周,檢測(cè)大鼠血、尿、便中尿酸水平,同時(shí)檢測(cè)肝、腎功能,并進(jìn)行組織學(xué)病理切片觀察。結(jié)果氧嗪酸鉀與乙胺丁醇聯(lián)合造模組大鼠血尿酸出現(xiàn)先升高后降低的現(xiàn)象,而氧嗪酸鉀與吡嗪酰胺聯(lián)合造模組在連續(xù)給藥期間血尿酸和尿液中尿酸升高平穩(wěn)。兩種方法對(duì)肝臟、腎臟均無(wú)明顯損害。結(jié)論氧嗪酸鉀與吡嗪酰胺聯(lián)合能有效建立尿酸排泄不良型高尿酸血癥大鼠模型,且模型穩(wěn)定性好,其發(fā)病過(guò)程更符合臨床尿酸排泄不良型高尿酸血癥的病程。
[Abstract]:Objective to explore the method of constructing reliable uric acid excretion adverse type hyperuricemia rat model, and lay the foundation for the optimization of treatment and pathogenesis of hyperuricemia uric acid excretion adverse type. Method using potassium oxonate (300 mg/kg) and pyrazinamide (300 mg/kg), ethambutol (250 mg/kg) combined with continuous administration model. 1 weeks, 3 weeks, 5 weeks, rats were detected in blood, urine, feces and uric acid, renal function, and detection of liver histological pathological observation. The potassium oxonate and ethambutol combined model rats serum uric acid increased first and then decreased, while the potassium oxonate and combined with pyrazinamide model group continuously during the administration of blood uric acid and urine uric acid increased steadily. The two methods on the liver and kidneys were no significant damage. Conclusion potassium oxonate combined with pyrazinamide can effectively establish uric acid excretion bad high uric acid in the blood The model of the rat, and the model is stable, is more consistent with the course of hyperuricemia in clinical uric acid excretion.
