人宮頸癌SiHa細(xì)胞荷瘤裸鼠模型的建立及鑒定

【摘要】:目的:建立人宮頸癌SiHa細(xì)胞裸小鼠體表荷瘤模型,并對(duì)其成瘤特性進(jìn)行分析和鑒定。方法:將5×10~6/mL、1×10~7/mL及5×10~7/mL 3個(gè)濃度的SiHa細(xì)胞懸液分別在3組裸小鼠背部右腋近上肢皮下接種,建立人宮頸癌裸小鼠體表荷瘤模型,觀察荷瘤裸鼠的成瘤率并測(cè)量腫瘤的大小,5周后取腫瘤組織經(jīng)病理學(xué)方法觀察其組織學(xué)特性,并采用PCR及免疫組織化學(xué)方法對(duì)腫瘤組織的HPV16E7 DNA表達(dá)進(jìn)行檢測(cè)。結(jié)果:3組裸小鼠接種SiHa細(xì)胞的成瘤率均為100%,5周后腫瘤大小分別達(dá)到(77.29±28.34)mm~3、(178.91±61.55)mm~3和(305.11±12.62)mm~3。3個(gè)濃度組間腫瘤體積大小差異有統(tǒng)計(jì)學(xué)意義(P0.001)。病理學(xué)大體觀察可見SiHa腫瘤的生長均以局部浸潤為主;組織切片觀察新生腫瘤細(xì)胞特點(diǎn)呈現(xiàn)體積大、核大深染及易見核分裂相的病理性特征;SiHa新生腫瘤組織經(jīng)PCR檢測(cè)證實(shí)了HPV16E7 DNA陽性,并經(jīng)免疫組織化學(xué)檢測(cè)證實(shí)了HPV16E7蛋白表達(dá)陽性。結(jié)論:成功建立了SiHa細(xì)胞荷瘤裸鼠模型,為人宮頸癌腫瘤靶向治療及生物學(xué)特性研究提供了理想的動(dòng)物模型。
[Abstract]:Objective: to establish a nude mouse model of human cervical cancer SiHa cells and to analyze and identify the tumorigenic characteristics of nude mice. Methods: 5 脳 10 ~ (6) / mL of human cervical cancer cells were used. Three concentrations of SiHa cell suspension of 1 脳 10 ~ (7) / mL and 5 脳 10 ~ (7) / mL were inoculated subcutaneously in the right axillary proximal upper extremity of three groups of nude mice to establish the model of human cervical carcinoma nude mice. The tumorigenesis rate of nude mice bearing tumor was observed and the tumor size was measured 5 weeks later, the histological characteristics of tumor tissue were observed by pathological method. PCR and immunohistochemical methods were used to detect the expression of HPV16E7 DNA in tumor tissue. Results the tumorigenic rate of the nude mice inoculated with SiHa cells in the three groups was 100%. After 5 weeks, the tumor size reached 77.29 鹵28.34 mm ~ (-3) respectively. There was significant difference in tumor volume size between 178.91 鹵61.55mm3 and 305.11 鹵12.62mm3 groups (P 0.001). Gross pathological observation showed that the growth of SiHa tumors was mainly local invasion. The histopathological features of neoplastic cells were large size, large nuclear staining and easy to see mitotic phase. HPV16E7 DNA was confirmed by PCR in the neoplasm of SiHa. The expression of HPV16E7 protein was confirmed by immunohistochemistry. Conclusion: the model of nude mice bearing tumor of SiHa cells was successfully established. Targeted therapy and biological characteristics of human cervical cancer provide an ideal animal model.
