體外光化學(xué)療法處理的調(diào)節(jié)性樹(shù)突細(xì)胞對(duì)T細(xì)胞增殖的影響

【摘要】:目的探討體外光化學(xué)療法處理的調(diào)節(jié)性樹(shù)突細(xì)胞對(duì)T細(xì)胞增殖的影響。方法獲取人外周血單個(gè)核細(xì)胞(PBMC),利用白介素-4重組人粒細(xì)、巨噬細(xì)胞集落刺激因子誘導(dǎo)生成未成熟的樹(shù)突細(xì)胞(im DC)。分離獲取人脾臟淋巴細(xì)胞(SP),經(jīng)光化學(xué)療法(PUVA)處理后獲得PUVA-SP;將im DC與PUVA-SP共培養(yǎng)得到體外光化學(xué)治療性樹(shù)突細(xì)胞(ecp DC);將im DC與SP共培養(yǎng)得到SPDC;im DC加入10ng/ml的脂多糖(LPS),培養(yǎng)1d后得到DC。收集4組細(xì)胞,檢測(cè)其表面抗體CD11c、CD83、CD86的表達(dá)。采用混合淋巴細(xì)胞培養(yǎng)的方法檢測(cè)吞噬了供者凋亡的脾淋巴細(xì)胞后的受者im DC對(duì)受者T細(xì)胞增殖的影響。結(jié)果經(jīng)PUVA處理后細(xì)胞早期凋亡率為91.33%。表面分子CD83、CD86在ecp DC的陽(yáng)性表達(dá)率為分別為22.83%±5.26%、22.06%±4.37%,與在im DC的陽(yáng)性表達(dá)率(分別為15.06%±0.59%、15.19%±1.83%)相近(P0.05),但明顯低于在DC的陽(yáng)性表達(dá)率(分別為99.79%±0.36%、99.85%±0.19%),差異有統(tǒng)計(jì)學(xué)意義(P0.01)。吞噬了供者凋亡的脾淋巴細(xì)胞后的受者im DC細(xì)胞可明顯抑制受者T細(xì)胞的增殖。結(jié)論體外光化學(xué)療法誘導(dǎo)的凋亡脾淋巴細(xì)胞可抑制樹(shù)突細(xì)胞的成熟,并可抑制受者T淋巴細(xì)胞的增殖。
[Abstract]:Objective to investigate the in vitro photodynamic therapy treatment of regulatory dendritic cells on the proliferation of T cell. Methods of obtaining human peripheral blood mononuclear cells (PBMC), using recombinant human interleukin -4 granule, macrophage colony stimulating factor induced the formation of immature dendritic cells (im DC). Isolated from human spleen lymphocytes (SP) photodynamic therapy (PUVA), was obtained after the treatment of PUVA-SP; Im DC and PUVA-SP were cultured in vitro by photochemical treatment of dendritic cells (ECP DC); Im DC co cultured with SP SPDC; Im DC 10ng/ml with lipopolysaccharide (LPS), after 1D DC. collected from 4 groups of cells, the detection of surface CD11c CD83, CD86 antibody, expression. By using the method of mixed lymphocyte culture test swallowed the effect of donor apoptotic splenocytes after recipient im DC on the proliferation of T cells. Results after treatment of PUVA cells early apoptosis rate of 91.33%. surface Molecular CD83, CD86 positive expression rate of DC in ECP were 22.83% + 5.26%, 22.06% + 4.37%, and the positive expression rate of DC in im (15.06% + 0.59%, 15.19% + 1.83%) similar to (P0.05), but was significantly lower than that in the positive expression rate of DC (99.79% + 0.36%, 99.85% + 0.19%), the difference was statistically significant (P0.01). The phagocytosis of donor spleen lymphocyte apoptosis after IM infected DC cells can significantly inhibited the proliferation of T cell apoptosis induced by splenic lymphocytes. Conclusion extracorporeal photochemotherapy can inhibit the maturation of dendritic cells, and inhibited the proliferation of T lymphocytes.
