蛋雞肝臟功能和DNA甲基化與產(chǎn)蛋率的關(guān)系

【摘要】:為研究產(chǎn)蛋后期產(chǎn)蛋率與肝臟功能及肝臟全基因組甲基化水平的關(guān)系,以海蘭褐蛋雞為試驗材料,對不同周齡、產(chǎn)蛋率蛋雞的肝功能指標(biāo)和全基因組甲基化情況進行了觀察分析。肝臟功能指標(biāo)包括脂肪代謝、氧化應(yīng)激、內(nèi)質(zhì)網(wǎng)應(yīng)激、免疫應(yīng)激這四個方面,肝臟全基因組甲基化情況主要從全基因組DNA甲基化圖譜、全基因組甲基化水平、差異甲基化基因、GO注釋、KEGG通路富集等方面進行了觀察分析。試驗一,不同周齡蛋雞的肝臟功能研究。根據(jù)蛋雞不同的周齡(25w和85w),選取16只高產(chǎn)蛋率的海蘭褐蛋雞分成兩個組:25w高產(chǎn)蛋率組、85w高產(chǎn)蛋率組。結(jié)果顯示,這2組蛋雞在血清乳酸脫氫酶、堿性磷酸酶、谷草轉(zhuǎn)氨酶、總蛋白、血清白蛋白、甘油三酯、總膽固醇、谷丙轉(zhuǎn)氨酶水平上沒有差異,85W蛋雞血清葡萄糖水平顯著升高(P0.05),SOD活性顯著降低(P0.05)。試驗二,不同產(chǎn)蛋率蛋雞的肝臟功能研究。挑選85周齡蛋雞不同的產(chǎn)蛋率(100%、70-80%、30-50%)共計24只海蘭褐蛋雞分成三個組:高產(chǎn)蛋率組、中等產(chǎn)蛋率組、低產(chǎn)蛋率組。結(jié)果表明,低產(chǎn)蛋率組蛋雞血清堿性磷酸酶有下降趨勢(P=0.0572)、葡萄糖水平有下降趨勢(P=0.0832),中產(chǎn)蛋率組蛋雞相比其他兩組肝臟CAT活性顯著降低(P0.05),蛋白質(zhì)羰基活性顯著升高(P0.05),ACC、FAS mRNA表達顯著降低(P0.05),85W-middle組蛋雞肝臟LXR mRNA表達顯著升高(P0.05),SOD1 mRNA表達有降低趨勢(P=0.0932),GPx1 mRNA表達有升高趨勢(P=0.0911)。低產(chǎn)蛋率組蛋雞肝臟甘油三酯含量顯著升高(P0.05),TNF-αmRNA表達顯著降低(P0.05)。試驗三,不同產(chǎn)蛋率蛋雞的肝臟甲基化研究。挑選70周齡蛋雞不同產(chǎn)蛋率(100%、40-60%),16只海蘭褐蛋雞分成兩個組:高產(chǎn)蛋率組、低產(chǎn)蛋率組。采用亞硫酸氫鹽高通量測序技術(shù),分析其全基因組甲基化。全基因組甲基化水平分別是56.98%、57.75%,不同染色體中C的甲基化水平、不同基因區(qū)域上各類型C甲基化水平、不同基因原件區(qū)域上各類型C甲基化水平結(jié)果中,低產(chǎn)蛋率組均高于高產(chǎn)蛋率組。由DMR(差異甲基化區(qū)域)得差異基因共3155,其中上調(diào)基因2172,下調(diào)基因983,低產(chǎn)蛋率組顯著性上調(diào)基因遠遠多于下調(diào)基因。在upstream區(qū)域和CDS區(qū)域,功能富集分析得到顯著富集GO分類注釋18條(P0.05),得到顯著富集pathway共三條(P0.05):嘌呤代謝、磷酸戊糖途徑、甘油磷脂代謝。低產(chǎn)蛋率組(low laying rate)蛋雞肝臟ATIC基因甲基化CDS區(qū)域上調(diào),其表達水平顯著升高(P0.05);低產(chǎn)蛋率組(low laying rate)蛋雞肝臟ALDOB基因甲基化CDS區(qū)域上調(diào),其表達水平顯著升高(P0.05)。低產(chǎn)蛋率組蛋雞肝臟LCPT1表達水平顯著升高(P0.05),LXR表達水平顯著升高(P0.05),ME1基因表達水平有降低趨勢(P=0.0656)。綜合以上結(jié)果,不同周齡產(chǎn)蛋率相近蛋雞肝功能指標(biāo)沒有明顯差別,同周齡不同產(chǎn)蛋率蛋雞間肝功能及肝臟全基因組甲基化存在明顯差異。
[Abstract]:In order to study the relationship between egg laying rate and liver function and the whole genome methylation level of liver, Hailan brown laying hens were used as experimental materials.The indexes of liver function and the whole genome methylation of laying hens were observed and analyzed.The liver function indexes include fat metabolism, oxidative stress, endoplasmic reticulum stress and immune stress. The whole genome methylation of liver is mainly based on the whole genome DNA methylation map and the whole genome methylation level.The enrichment of KEGG pathway of differential methylation gene (go) was observed and analyzed.Experiment 1: liver function of layers at different weeks of age.According to different weeks of age, 16 Hailan brown laying hens with high laying rate were divided into two groups: 1: 25 w high laying rate group and 85 w high laying rate group.The results showed that the two groups of layers in the serum lactate dehydrogenase, alkaline phosphatase, glutamic oxaloacetic transaminase, total protein, serum albumin, triglyceride, total cholesterol,There was no difference in the level of glutamate-pyruvate aminotransferase (alt). The serum glucose level of 85 W laying hens was significantly increased and the SOD activity of P0.05 was significantly decreased.The second experiment was to study the liver function of laying hens with different laying rates.A total of 24 Hailan brown layers were divided into three groups: high laying rate group, medium laying rate group and low laying rate group.The results show thatIn low laying rate group, the content of triglyceride in liver increased significantly, and the expression of TNF- 偽 mRNA decreased significantly.Experiment 3. Study on liver methylation of laying hens with different laying rates.Sixteen Hailan brown layers were divided into two groups: high laying rate group and low laying rate group.The whole genome methylation of H2SO4 was analyzed by high throughput sequencing technique.The whole genome methylation levels were 56.98 and 57.75, respectively. The levels of C methylation in different chromosomes, in different gene regions, in all types of C methylation levels in different gene regions, in different gene original regions, in all types of C methylation levels,The low laying rate group was higher than the high laying rate group.A total of 3155 differentially expressed genes were obtained from DMRs (differential methylation region). Among them, 2172 genes were up-regulated and 983down-regulated genes were down-regulated. In the low laying rate group, the up-regulated genes were much more than the down-regulated genes.In the upstream region and CDS region, the functional enrichment analysis showed that there were significant enrichment of P0.05 in 18 Go classification notes, and a significant enrichment of P0.05 in pathway: purine metabolism, pentose phosphate pathway, glycerol phospholipid metabolism.In low laying rate group, the methylation CDS region of ATIC gene in the liver of laying hens was upregulated, and its expression level was significantly increased (P 0.05), and the methylated CDS region of ALDOB gene in liver was up-regulated in low laying rate group (P 0.05).In low laying rate group, the expression level of LCPT1 in liver of laying hens increased significantly, and the expression level of LXR of P0.05An increased significantly. There was a tendency to decrease the expression level of P0.05AME1 gene in laying hens with a tendency of decreasing P0.0656.Based on the above results, there was no significant difference in the liver function index of the layers with the similar laying rate at different weeks, but there were significant differences in the liver function and the whole liver genome methylation among the layers with different laying rates at the same age.
