【摘要】:鈣磷作為蛋雞骨骼和蛋殼的重要組成成分,與蛋雞蛋殼品質(zhì)密切相關(guān)。隨著產(chǎn)蛋期的延長,由于蛋雞鈣磷代謝紊亂而導(dǎo)致蛋殼品質(zhì)下降是制約現(xiàn)代蛋雞集約化養(yǎng)殖業(yè)發(fā)展的主要問題之一。成纖維細胞生長因子23(FGF23)屬于成纖維細胞生長因子家族,主要由骨細胞和成骨細胞合成分泌,是新近發(fā)現(xiàn)的體內(nèi)調(diào)節(jié)鈣磷代謝的重要因子,F(xiàn)有文獻表明,FGF23可能就是哺乳動物體內(nèi)腸道-腎臟-甲狀旁腺-骨骼之間調(diào)控機體鈣磷代謝的偶聯(lián)因子。一般認為FGF23主要在腎臟與腎小管上皮細胞膜上的Klotho-FGFRs(成纖維生長因子受體)復(fù)合物特異性結(jié)合后通過激活MAPK信號通路調(diào)節(jié)腎小管對鈣磷的重新收。但目前,FGF23-Klotho在家禽體內(nèi)的作用機制尚不清楚。本文通過RT-PCR方法檢測了FGF23和KlothomRNA在產(chǎn)蛋雞胸肌、骨骼肌、腎臟、心臟、肝臟、骨關(guān)節(jié)、骨、甲狀旁腺、腎上腺、蛋殼腺、卵巢、下丘腦、垂體和十二指腸組織中的表達情況;利用實時熒光定量RT-PCR法檢測了不同產(chǎn)蛋期骨組織中FGF23 mRNA及腎臟中FGF23-Klotho作用相關(guān)因子(Klotho、FGFR1、FGFR3、FGFR4、TRPV5、TRPV6、NCX1、PMCA1b、NaPi-Ⅱa、CaBP-D28k和Cyp24a1)的表達水平;常規(guī)方法檢測蛋殼品質(zhì),探討不同產(chǎn)蛋期腎臟中FGF23-Klotho作用因子表達變化研究,以期為今后進一步研究FGF23和Klotho在蛋雞生產(chǎn)中的作用提供前期基礎(chǔ)。試驗結(jié)果證明:1.蛋雞骨關(guān)節(jié)、卵巢和肝臟組織中存在FGF23mRNA表達;Klotho在蛋雞垂體、心臟、十二指腸、卵巢、蛋殼腺、腎臟、腎上腺、甲狀旁腺和下丘腦等組織中均有廣泛表達。2.產(chǎn)蛋前期和產(chǎn)蛋高峰期蛋雞骨關(guān)節(jié)FGF23 mRNA表達水平顯著高于產(chǎn)蛋后期;產(chǎn)蛋高峰期腎臟中FGFR1、FGFR3、FGFR4、CaBP-D28k及PMCA1b mRNA表達水平顯著高于產(chǎn)蛋前期;產(chǎn)蛋后期TRPV5和Cyp24a1 mRNA表達水平顯著低于產(chǎn)蛋前期。3.產(chǎn)蛋后期的蛋殼厚度、蛋殼強度以及蛋殼比例均顯著低于產(chǎn)蛋前期和產(chǎn)蛋高峰期。本試驗結(jié)果表明,產(chǎn)蛋后期蛋殼品質(zhì)的降低與腎臟對鈣重吸收能力下降密切相關(guān)。
[Abstract]:As an important component of the skeleton and eggshell of laying hens, calcium and phosphorus are closely related to the quality of egg and egg shell. With the prolongation of laying period, the quality of eggshell resulting from the disorder of calcium and phosphorus in laying hens is one of the main problems that restrict the development of modern laying hens. Fibroblast growth factor 23 (FGF23) is a fibroblast growth factor family, mainly composed of bone cells and osteoblasts. It is a newly discovered important factor regulating calcium and phosphorus metabolism in vivo. The existing literature indicates that FGF23 may be the coupling factor that regulates the metabolism of calcium and phosphorus between the intestinal - kidney - parathyroid - bone in the mammalian body. It is generally believed that FGF23 specifically regulates the recovery of calcium and phosphorus in renal tubules by activating MAPK signaling pathway after binding to the Klotho-FGFRs (fibroblast growth factor receptor) complex on the membrane of kidney and renal tubular epithelial cell membrane. But at present, the mechanism of FGF23-Klotho in poultry is not clear. This paper examined the expression of FGF23 and KlothomRNA in laying hens muscle, skeletal muscle, kidney, heart, liver, bone, bone, adrenal gland, parathyroid, eggshell, ovary, hypothalamus, pituitary and duodenal tissues were detected by RT-PCR method; the role of FGF23 and mRNA related factor FGF23-Klotho in kidney tissue of different laying period by using real time fluorescence quantitative RT-PCR (Klotho, FGFR1, FGFR3, FGFR4, TRPV5, TRPV6, NCX1, PMCA1b, a, CaBP-D28k and NaPi- II Cyp24a1) expression level; conventional method for detection of eggshell quality, to investigate the changes of the function of FGF23-Klotho in the kidney of expression of different laying period, provide a basis for further research on FGF23 and Klotho in laying hens in order to. The results showed that: 1., there was FGF23mRNA expression in bone, joint, ovary and liver tissues of the laying hens. Klotho was widely expressed in the tissues of layers, such as pituitary, heart, duodenum, ovary, eggshell gland, kidney, adrenal gland, parathyroid gland and hypothalamus. 2. egg laying hens and early osteoarthritis FGF23 mRNA expression level was significantly higher than that of egg laying; the expression of FGFR1, FGFR3, FGFR4 egg, CaBP-D28k PMCA1b and mRNA peak in kidney were significantly higher than that of the early laying period; laying TRPV5 and Cyp24a1 mRNA expression level was significantly lower than that of the Early Laying period. 3. the thickness of eggshell, the strength of eggshell and the proportion of eggshell were significantly lower than the pre - laying period and the peak period of egg production. The results showed that the decrease of eggshell quality was closely related to the decrease of renal calcium reabsorption capacity in the later period of laying.
