目的胰腺癌是一種全球性的重要健康問題,尤其是在中國,胰腺癌是死亡率最高的惡性腫瘤之一。胰腺癌轉(zhuǎn)移和術(shù)后復(fù)發(fā)嚴(yán)重影響患者的療效和生存質(zhì)量,是導(dǎo)致胰腺癌患者死亡的主要原因。在過去十年中,盡管手術(shù)方法和輔助治療手段有了長足的進(jìn)步,胰腺癌死亡率依然居高不下。一個很重要的原因是大多數(shù)患者就診時已經(jīng)達(dá)到晚期,且伴有遠(yuǎn)處轉(zhuǎn)移。因此,深入研究胰腺癌轉(zhuǎn)移的機(jī)制,尋找胰腺癌轉(zhuǎn)移的早期診斷與阻斷治療靶點尤為重要。本研究的目的在于探討膠原蛋白VIα1(COL6A1)促進(jìn)胰腺癌轉(zhuǎn)移的作用及臨床意義。方法以胰腺癌細(xì)胞株BxPC-3為親代,反復(fù)穿越Transwell小室8次,構(gòu)建新型高轉(zhuǎn)移細(xì)胞模型BxPC-M8。通過劃痕、遷移、侵襲等實驗比較BxPC-3與BxPC-M8細(xì)胞體外的遷移和侵襲能力。應(yīng)用基因組測序與實時定量PCR(qRT-PCR)驗證找出調(diào)控胰腺癌轉(zhuǎn)移的潛在基因——COL6A1。通過siRNA敲低實驗研究COL6A1對胰腺癌細(xì)胞轉(zhuǎn)移能力的影響。同時,采用WB檢測敲低COL6A1后對胰腺癌細(xì)胞上皮間質(zhì)轉(zhuǎn)化(EMT)相關(guān)蛋白表達(dá)的影響。最后利用組織芯片檢測技術(shù),分析COL6A1表達(dá)異常與胰腺癌患者臨床病理特...
