翼狀胬肉的體外細胞模型建立和分化誘導治療研究

目的:翼狀胬肉是一種侵襲性增生,是角膜上皮細胞過度增殖的結(jié)果。分化治療近年來引起了人們的廣泛關注。本研究的目的是從翼狀胬肉中鑒定干細胞,并檢測翼狀胬肉干細胞的分化策略。分化療法的概念來源于激素、細胞因子或不同分化劑可以促進分化,并且可以不可逆地改變細胞表型。本文簡要地介紹了幾篇文獻和研究,以證明目前的目標,即分化治療在治療多種臨床疾病中能夠發(fā)揮關鍵和有前途的作用。方法:應用體外原代培養(yǎng)技術(shù)獲得胬肉干細胞,應用免疫熒光染色法研究培養(yǎng)的翼狀胬肉干細胞的干細胞標志物和多向分化潛能等干細胞特性。誘導分化后利用克隆形成實驗和MTT法檢測細胞活力,檢測和評價分化治療策略在胬肉治療中的應用潛能。結(jié)果:翼狀胬肉干細胞可在體外進行持續(xù)的培養(yǎng)和擴增。它們表達了多能干細胞標記物,可在體外誘導分化為神經(jīng)元細胞、骨細胞和脂肪細胞。經(jīng)誘導試驗處理后,翼狀胬肉干細胞球表達干細胞標志物和上皮-間充質(zhì)轉(zhuǎn)化標志物。誘導分化后,翼狀胬肉干細胞的增殖和存活能力受到抑制,提示分化誘導策略在翼狀胬肉治療中有廣闊的應用前景。結(jié)論:翼狀胬肉來源的細胞表達多種干細胞標志物并且具備多向誘導分化潛能。胬肉干細胞在體外經(jīng)過誘導培養(yǎng)基處理后,...
