MicroRNA-539在胰腺癌中的表達及其調(diào)控機制研究

胰腺癌是高度惡性的消化道腫瘤,被發(fā)現(xiàn)時患者往往處于癌癥晚期,只有極少部分患者早期能得到診斷,并進行下一步的根治性手術(shù)治療。在臨床上目前還沒有敏感性高特異性強的診斷標(biāo)志物來對胰腺癌進行早期診斷,同時也沒有一個有效的手段來延長胰腺癌患者的生存期。胰腺癌患者僅不到20%可行手術(shù),對于無法手術(shù)的患者,介入與常規(guī)的細胞毒性化學(xué)治療效果不佳,尋找新的治療手段已迫在眉睫。在人類的惡性疾病中,microRNA出現(xiàn)了不同程度的表達的異常,這些結(jié)果顯示其在腫瘤發(fā)生發(fā)展過程中可能起著重要作用。本研究擬采用相關(guān)的生物學(xué)技術(shù)及方法來初步探討miR-539在胰腺癌中的表達及其對胰腺癌生物學(xué)行為影響的調(diào)控機制。第一部分microRNA-539表達下調(diào)提示胰腺癌患者預(yù)后不良本研究擬分析miR-539表達水平與胰腺癌患者臨床病理特征及預(yù)后的相關(guān)性。我們用RT-qPCR技術(shù)檢測了 56例胰腺癌患者中miR-539的表達水平,并將它與癌旁組織進行比較,我們發(fā)現(xiàn),胰腺癌患者中miR-539表達水平顯著下調(diào)。miR-539的表達水平與淋巴結(jié)轉(zhuǎn)移(有VS無)、臨床分期(Ⅲ-Ⅳ與Ⅰ-Ⅱ)、CA 199(≥200與<200)和...
