優(yōu)化納米藥物設(shè)計增強其抗腫瘤效果的研究

腫瘤納米藥物可以通過多種方式攜載傳統(tǒng)藥物,增加其體內(nèi)穩(wěn)定性和延長藥物血液半衰期,提高其生物利用度。納米藥物可以通過其特有的EPR效應(yīng)在腫瘤部位高效富集,提高了藥物在腫瘤部位的濃度,同時減少了在正常部位的藥物濃度,從而提高了腫瘤的治療效果,降低了藥物的副作用。這些優(yōu)勢使得納米藥物在腫瘤治療中突顯出了極大的應(yīng)用潛力。目前,已經(jīng)有部分納米藥物進入到癌癥臨床試驗階段或者上市。然而,大部分癌癥納米藥物臨床試驗結(jié)果并不樂觀,能最終進入臨床的納米藥物的比例極少,這也是目前基于納米技術(shù)的腫瘤療法并沒有得到廣泛應(yīng)用的原因。因此,如何進一步提高腫瘤納米藥物的治療效果仍然是亟待解決的關(guān)鍵科學(xué)問題。納米藥物具有多種特性(如尺寸、電荷、親疏水性、表面化學(xué)等),這些特性顯著影響其與生物體的相互作用及其體內(nèi)命運。因此,研究納米藥物的特性與其生物學(xué)行為之間的相關(guān)性,是建立納米藥物設(shè)計指南和提高腫瘤治療效果的有效手段。另一方面,放療是目前腫瘤治療的主要手段之一。在臨床上,超過一半的癌癥患者會接受放療。然而,目前放療在腫瘤治療中仍然會受到一些因素的限制(放療耐受)。此外,盡管腫瘤細胞由于具有較高的有絲分裂活性和DNA損傷...
