三株地衣內(nèi)生真菌的化學(xué)成分研究

【摘要】:內(nèi)生菌是指存活于健康有機(jī)體組織內(nèi)部而不引起任何直接或明顯負(fù)面影響的微生物,主要包括真菌,細(xì)菌和放線菌等。內(nèi)生真菌被研究最多,已經(jīng)被認(rèn)為是可持續(xù)的、生物活性多樣的和結(jié)構(gòu)新穎的先導(dǎo)化合物的重要來源。地衣內(nèi)生真菌(Endolichenic fungi)是指生活在無癥狀的地衣菌體內(nèi)部的真菌,類似于植物內(nèi)生真菌寄生于健康植物組織。由于地衣菌藻共生等特殊性,地衣內(nèi)生真菌經(jīng)常被忽略,然而自從2007年第一次報道它們的次生代謝產(chǎn)物,其化學(xué)成分的研究正越來越引起學(xué)者的注意。 本文對三株地衣內(nèi)生真菌(Phaeosphaeria sp.、Massarina sp.、Cucurbitaria sp.)的化學(xué)成分進(jìn)行了系統(tǒng)研究,采用常規(guī)的硅膠柱層析、葡聚糖凝膠SephadexLH-20柱層析以及高效液相色譜等分離手段共分離鑒定了37個化合物,包括13個傒醌,3個三聯(lián)苯,5個芳香類,2個脂肪酸,8個甾體和6個其它類化合物,其中新化合物12個,包括7個花醌:phaeosphaerin A-G (1-7);1個脂肪酸:(11S,12S,13R)-11,13-dihydroxy-12-methyltetradecanoic acid (19);1個α,β-不飽和酮類化合物:5-hydroxy-2,3-dimethylcyclohex-2-enone (30);以及3個三聯(lián)苯:cucurbitarins A-C (31-33)。利用各種現(xiàn)代波譜技術(shù)包括質(zhì)譜(MS)、核磁共振譜(NMR),紅外波譜(IR)和紫外波譜(UV),結(jié)合含時密度泛函理論(TDDFT) CD計算等方法闡明了新結(jié)構(gòu)化合物的平面與立體結(jié)構(gòu)。同時,對分離得到的部分化合物進(jìn)行了針對不同腫瘤細(xì)胞株的細(xì)胞毒活性評價以及真菌白色念珠菌的抗菌活性評價,花醌類化合物富集于腫瘤細(xì)胞溶酶體顯示較強(qiáng)的抗PC3, DU145和LNCaP前列腺癌細(xì)胞系的生物活性。
[Abstract]:Endophytic bacteria (endophytes) are microorganisms that survive in healthy organisms without any direct or obvious negative effects, including fungi, bacteria and actinomycetes. Endophytic fungi have been widely studied and considered to be an important source of sustainable, bioactive and novel precursor compounds. Lichen endophytic fungus (Endolichenic fungi) is a kind of fungus living inside the asymptomatic lichens, which is similar to plant endophytic fungi parasitic on healthy plant tissues. Because of the particularity of lichenium-algae symbiosis endophytic fungi are often neglected. However since the first report of their secondary metabolites in 2007 the study of their chemical constituents has attracted more and more attention. Three strains of endophytic fungi (Phaeosphaeria sp.,Massarina sp.,Cucurbitaria sp.) from lichen were studied in this paper. 37 compounds were isolated and identified by conventional silica gel column chromatography, dextran gel SephadexLH-20 column chromatography and high performance liquid chromatography. It includes 13 quinones, 3 triphenyls, 5 aromatics, 2 fatty acids, 8 steroids and 6 other compounds, of which 12 are new compounds. It consists of seven quinones: phaeosphaerin A-G (1-7); one fatty acid: (11Sn12Sn13R) -1113-dihydroxy-12-methyltetradecanoic acid (19); one 偽, 尾 -unsaturated ketones: 5-hydroxy-23-dimethylcyclohex-2-enone (30); and three triphenyls: cucurbitarins A-C (31-33). The plane and solid structures of the new compounds were elucidated by various modern spectral techniques including (MS), NMR (NMR), IR (IR) and UV (UV), combined with time-dependent density functional theory (TDDFT) CD). At the same time, some of the compounds were evaluated for the cytotoxic activity of different tumor cell lines and the antibacterial activity of fungi Candida albicans. Anthraquinones enriched in lysosomes of tumor cells showed strong biological activity against PC3, DU145 and LNCaP prostate cancer cell lines.
