七氟醚后處理對微管結(jié)構(gòu)及心肌保護(hù)效果的影響

【摘要】:目的:探討七氟醚后處理對小鼠心肌缺氧/復(fù)氧損傷時微管結(jié)構(gòu)變化的影響,以及心肌實(shí)施七氟醚后處理的心肌保護(hù)效果的可行性分析。方法:首先培養(yǎng)小鼠乳鼠原代心肌細(xì)胞,建立缺氧/復(fù)氧損傷模型,將貼壁的心肌細(xì)胞隨機(jī)分為5組:假手術(shù)組(sham組)、缺氧/復(fù)氧損傷(H/R組)、七氟醚后處理組(SPostC組)、七氟醚后處理組+紫杉醇組(SPostC+toxl組)、七氟醚后處理組+秋水仙素組(SPostC+Col組),免疫熒光染色后用共聚焦顯微鏡觀察細(xì)胞微管分布、形態(tài)變化以及比較微管蛋白熒光強(qiáng)度。然后將C57小鼠(20-30g)40只,制備在體心肌缺血再灌注損傷(I/R)模型,隨機(jī)分組同上;分別于缺血前、再灌組30min、120min后觀察收縮壓、心率等血流動力學(xué)指標(biāo),于再灌組120min比較各組左心室心梗面積;然后用蛋白質(zhì)印跡法檢測(western blot)心肌α微管蛋白的表達(dá)。結(jié)果:與缺氧/復(fù)氧損傷組(H/R)比較,七氟醚后處理組微管結(jié)構(gòu)的損傷有所減輕,且微管α蛋白免疫熒光蛋白強(qiáng)度增加(P0.05);且能明顯減少小鼠左心室心肌梗死面積(P0.05);在再灌組120min時,七氟醚后處理組血壓升高(P0.05)、心率減慢(P0.05);western blot也證實(shí)與缺血再灌注組比較,七氟醚后處理組能明顯增加α-微管蛋白的表達(dá)(P0.05)。結(jié)論:七氟醚后處理可以明顯減輕缺血再灌注所致的微管結(jié)構(gòu)的損傷,且具有一定的心肌保護(hù)作用,為微管相關(guān)的心肌保護(hù)提供一定的理論依據(jù)。
[Abstract]:Objective: to investigate the effect of sevoflurane post-treatment on the changes of microtubule structure during hypoxia / reoxygenation injury in mice and the feasibility analysis of myocardial protective effect of sevoflurane post-treatment. Methods: the primary cardiomyocytes of neonatal mice were cultured at first, and the model of hypoxia / reoxygenation injury was established. The adherent cardiomyocytes were randomly divided into 5 groups: sham operation group (sham group), hypoxia / reoxygenation injury group (H / R group), sevoflurane post-treatment group (SPostC group), sevoflurane post-treated paclitaxel group (SPostC toxl group) and sevoflurane post-treated colchicine group. The distribution of microtubules in SPostC Col group was observed by confocal microscope after immunofluorescence staining. Morphological changes and comparison of fluorescent intensity of tubulin. Then, 40 C57 mice (20-30g) were used to establish myocardial ischemia-reperfusion injury (I / R) model in vivo, and were randomly divided into the same group, systolic blood pressure, heart rate and other hemodynamic indexes were observed before ischemia and 30 min after reperfusion group. The area of left ventricular myocardial infarction (LVMI) was compared with that of reperfusion group (120min), and the expression of 偽 tubulin in (western blot) myocardium was detected by Western blotting. Results: compared with the hypoxia / reoxygenation group (H / R), the damage of microtubule structure in sevoflurane post-treated group was alleviated, and the intensity of microtubule 偽 protein immunofluorescence protein was increased (P0.05), and the myocardial infarction area of left ventricle in mice was significantly decreased (P0.05). In sevoflurane post-treatment group, the blood pressure was increased (P0.05), and the heart rate was decreased (P0.05). Western blot also confirmed that sevoflurane post-treated group could significantly increase the expression of 偽 -tubulin compared with the ischemia-reperfusion group (P0.05). Conclusion: sevoflurane post-treatment can obviously reduce the injury of microtubule structure induced by ischemia reperfusion and has certain myocardial protective effect, which provides a theoretical basis for microtubule-related myocardial protection.
