【摘要】：目的:觀察參芪補(bǔ)肺方對(duì)COPD大鼠KLF2、Nrf2、y-GCS蛋白和mRNA的影響。方法:將健康、清潔級(jí)雌性Wistar大鼠40只,隨機(jī)分為空白組、模型組、富露施組、中藥組。采用香煙煙霧熏制和經(jīng)鼻給予脂多糖法制作COPD大鼠模型。鏡下觀察大鼠肺組織病理形態(tài)學(xué)改變;RT-PCR方法檢測(cè)COPD大鼠肺組織中KLF2、Nrf2、Y-GCSmRNA的表達(dá);免疫組化方法檢測(cè)COPD大鼠肺組織中KLF2、Nrf2、Y-GCS蛋白的表達(dá)。結(jié)果:1.HE染色后鏡下觀察大鼠肺組織,發(fā)現(xiàn)模型組、富露施組、中藥組的肺組織改變與COPD病理改變相同。2.RT-PCR結(jié)果顯示:模型組、富露施組、中藥組大鼠肺組織KLF2、Nrf2、γ-GCS mRNA的表達(dá)高于空白組,經(jīng)統(tǒng)計(jì)學(xué)處理有明顯差異(P0.05);模型組大鼠肺組織KLF2、Nrf2、γ-GCSmRNA的表達(dá)明顯高于富露施組、中藥組,經(jīng)統(tǒng)計(jì)學(xué)處理有意義(P0.05)。3.免疫組化結(jié)果顯示:KLF2、Nrf2、Y-GCS蛋白見于肺泡、支氣管、血管內(nèi)皮細(xì)胞;模型組、富露施組、中藥組肺組織中KLF2、Nrf2、γ-GCS蛋白的表達(dá)明顯高于空白組,經(jīng)統(tǒng)計(jì)學(xué)處理有明顯差異(P0.05);富露施組、中藥組KLF2、Nrf2、γ-GCS蛋白的表達(dá)低于模型組經(jīng)統(tǒng)計(jì)學(xué)處理有差異(P0.05)。4.富露施組與中藥組的肺組織中KLF2、Nrf2、Y-GCS mRNA和蛋白的表達(dá)相近,無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:1.參芪補(bǔ)肺方可以減輕COPD大鼠肺組織的病理改變2.參芪補(bǔ)肺方可增加COPD大鼠肺組織中KLF2、Nrf2、γ-GCSmRNA和蛋白的表達(dá),增加組織的抗氧化能力。
[Abstract]:Objective: To observe the effect of Shenqi tonifying lung Recipe on KLF2, Nrf2, y-GCS protein and mRNA in COPD rats. Methods: 40 healthy and clean female Wistar rats were randomly divided into blank group, model group, rich exposure group, Chinese medicine group. The rat model of COPD was made with cigarette smoke fumigating and transnasal lipopolysaccharide. The pathological morphology of lung tissue was observed under the microscope. The expression of KLF2, Nrf2 and Y-GCSmRNA in lung tissue of COPD rats was detected by RT-PCR method, and the expression of KLF2, Nrf2 and Y-GCS protein in lung tissue of COPD rats was detected by immunohistochemistry. Results: the lung tissue of rats was observed under the microscope after 1.HE staining, and the model group, rich exposure group, and the same.2.RT-PCR knot in the lung tissue of the Chinese medicine group and COPD pathological changes were found. The results showed that the expression of KLF2, Nrf2 and -GCS mRNA in the lung tissue of the rats in the model group and Fu Lu Shi group was higher than that in the blank group (P0.05). The expression of KLF2, Nrf2, and gamma -GCSmRNA in the lung tissue of the model group was significantly higher than that in the rich exposed group. The results of statistical processing showed significant (P0.05).3. immuno histochemical results showed: KLF2, Nrf2, Y-GCS protein was found in alveolar, bronchial, vascular endothelial cells, model group, rich exposure group, the expression of KLF2, Nrf2, gamma -GCS protein in the lung tissue of the Chinese medicine group was significantly higher than that in the blank group (P0.05). The expression of KLF2, Nrf2, and gamma -GCS protein in the group of Fu Lu Shi group was lower than that of the model group (P0.05). 4. rich dew KLF2, Shi Group and Chinese medicine group in the lung tissues of Nrf2, Y-GCS and mRNA protein expression are similar, no statistical significance (P0.05). Conclusion: 1. Shenqi Bufei recipe can reduce rat lung tissue pathological changes of COPD 2. Shenqi Bufei recipe can increase KLF2 and COPD of lung tissues in rats in Nrf2, the expression of gamma -GCSmRNA and protein, increase the antioxidant capacity of the tissues.
