續(xù)骨丹對大鼠骨缺損模型血清BMP-2的影響

【摘要】:目的:運用Elisa、HE染色、X光檢測等技術(shù),觀察續(xù)骨丹外敷療法對大鼠脛骨大塊缺損模型血清骨形態(tài)發(fā)生蛋白-2(BMP-2)表達(dá)的影響,以及誘導(dǎo)骨缺損的成骨情況和最終骨修復(fù)的結(jié)果,初步揭示續(xù)骨丹促進(jìn)骨缺損修復(fù)、康復(fù)的機(jī)理。方法:將70只SD大鼠右側(cè)脛骨制備1個4mm缺損,用0.8mm克氏針行內(nèi)固定。造模1個月后將符合標(biāo)準(zhǔn)的大鼠隨機(jī)分為實驗組(藥物治療組)、對照組a(脈沖磁場治療組)、對照組b(面粉治療組)和空白組。分別在干預(yù)后的0、2、4周,對大鼠右脛骨進(jìn)行X線檢查,然后處死動物,收獲骨樣本及血清,做蘇木素-伊紅(HE)染色和血清BMP-2酶聯(lián)免疫檢測。結(jié)果:①干預(yù)0周時,實驗組、對照組a、對照組b和空白組大鼠血清BMP-2的含量沒有明顯差異;干預(yù)至第2周時,四組大鼠血清BMP-2的含量與干預(yù)前相比均下降,但組間差異均無統(tǒng)計學(xué)意義(P0.05);干預(yù)4周后,實驗組大鼠血清BMP-2的含量高于對照組a,差異有統(tǒng)計學(xué)意義(P0.05),實驗組大鼠血清BMP-2的含量高于對照組b和空白組,差異有統(tǒng)計學(xué)意義(P0.01),對照組a血清BMP-2含量明顯高于對照組b和空白組,差異有統(tǒng)計學(xué)意義(P<0.01),對照組b與空白組比較,無明顯差異(p0.05)。三組大鼠血清BMP-2的含量在干預(yù)0、2、4周后的變化趨勢是:實驗組、對照組a先降后升,對照組b和空白組一直下降。②干預(yù)至第4周,四組大鼠骨樣本HE染色結(jié)果顯示:實驗組大鼠中的4只和對照組a中的2只顯示橫斷面上骨組織基本完整,骨髓組織豐富或比較豐富,少量骨組織與骨髓組織交錯存在;空白組及對照組b大鼠染色均顯示骨組織不同程度的殘缺及骨髓組織稀少或欠豐富。③干預(yù)至第4周,四組大鼠骨缺損造模側(cè)肢體X光片顯示:實驗組5只大鼠有4只基本愈合,對照組a中5只大鼠有2只基本愈合,愈合脛骨X片顯示脛骨原缺損處骨皮質(zhì)連續(xù),未見骨折線存在;對照組b和空白組大鼠右脛骨均未完全愈合,X片顯示脛骨中下段骨皮質(zhì)不連,骨折線存在明顯,周圍未見明顯骨痂生長。結(jié)論:續(xù)骨丹外敷療法和脈沖電磁場療法均能在干預(yù)至第4周時使處于下降趨勢內(nèi)源性BMP-2重新高表達(dá),誘導(dǎo)骨修復(fù),促進(jìn)骨缺損的康復(fù)愈合。但續(xù)骨丹外敷療法在對BMP-2的調(diào)節(jié)作用上明顯優(yōu)于脈沖電磁場療法。
[Abstract]:Objective: to observe the effect of Xray on the expression of serum bone morphogenetic protein (BMP-2), bone morphogenetic protein-2 (BMP-2) in rats with large tibial defect, and to investigate the osteogenesis of bone defect and the result of bone repair.The mechanism of promoting bone defect repair and rehabilitation was revealed.Methods: one 4mm defect was prepared from the right tibia of 70 SD rats and was fixed with 0.8mm Kirschner needle.One month later, the rats who met the standard were randomly divided into experimental group (drug treatment group), control group (pulse magnetic field treatment group), control group B (flour treatment group) and blank group.The right tibia of the rats were examined by X-ray at 0 ~ 2 weeks after intervention, and then the animals were killed, bone samples and serum samples were harvested, hematoxylin-eosin) staining and serum BMP-2 enzyme-linked immunosorbent assay were performed.Results there was no significant difference in the content of serum BMP-2 between experimental group, control group, control group and blank group at 0 weeks after intervention by 1: 1, and by the second week of intervention, the content of serum BMP-2 in the four groups was lower than that before intervention.After 4 weeks of intervention, the content of serum BMP-2 in the experimental group was higher than that in the control group (P 0.05), and the content of serum BMP-2 in the experimental group was higher than that in the control group (b) and the blank group (P < 0.05).The level of serum BMP-2 in control group was significantly higher than that in control group and blank group (P < 0.01), but there was no significant difference between control group and blank group (P < 0.01).The change trend of serum BMP-2 content in the three groups after 4 weeks of intervention was as follows: in the experimental group, the control group a decreased first and then increased, the control group b and the blank group decreased 2. 2 until the 4th week.The results of HE staining in bone samples of the four groups showed that 4 rats in the experimental group and 2 rats in the control group a showed that the bone tissue on the cross section was basically intact, the bone marrow tissue was abundant or relatively abundant, and a small amount of bone tissue interlaced with the bone marrow tissue.In the blank group and the control group, the staining of b rats showed that the bone tissue was damaged to different degree and the bone marrow tissue was rare or deficient. 3.X ray films of the limbs of the four groups showed that 4 of the 5 rats in the experimental group were basically healed, 2 of the 5 rats in the control group were basically healed, and the X ray of the healed tibia showed the bone cortex of the original defect of the tibia was continuous.No fracture line was found in the control group b and the right tibia was not completely healed in the control group (b) and the right tibia in the blank group was not completely healed. The bone cortex of the middle and lower tibia was not connected, the fracture line was obvious, and there was no obvious callus growth around it.Conclusion: both external application and pulsed electromagnetic field therapy can reexpress endogenous BMP-2 in the descending trend, induce bone repair and promote the healing of bone defect.But the external application of Xiegudan was superior to pulse electromagnetic field therapy in regulating BMP-2.
