【摘要】：目的:探討新疆哈薩克族阿爾茨海默病中醫(yī)證型與ApoE基因多態(tài)性的相關(guān)性研究。方法:收集2010年6月-2016年11月在新疆阿勒泰地區(qū)完成AD流行病學(xué)調(diào)查的基礎(chǔ)上診斷哈薩克族AD患者及新疆神經(jīng)退行性疾病臨床大數(shù)據(jù)庫的AD患者共58例,對照組共79例,將病例組按中醫(yī)診斷標(biāo)準(zhǔn)及常用證候分型標(biāo)準(zhǔn)進(jìn)行分類,通過對各組人群ApoE基因型的檢測,比較其分布的差異性。結(jié)果:通過本次實驗研究,新疆哈薩克族人群兩組基因型分布在AD患者中,ApoEε2攜帶者比例明顯降低,ApoEε4攜帶者比例明顯升高;在正常人群中,ApoEε3攜帶者比例最高,ApoEε2次之,ApoEε4最低。兩組等位基因頻率有顯著差異性(P0.01)。中醫(yī)分型后的ApoE基因型在病例組和對照組中的分布,腎精虧虛證ApoEε3/ε4基因型與對照組相比其分布較高(P0.05);與其他證型相比較亦有差異性(P0.05)。同時,rs429358T/C位點突變基因頻率分布有統(tǒng)計學(xué)意義(P0.05),對于其它位點(rs769449、rs7412、rs405509)在現(xiàn)有的樣本數(shù)上沒有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:通過本課題的研究,發(fā)現(xiàn)新疆哈薩克族阿爾茨海默病中醫(yī)證型與ApoE基因多態(tài)性之間有相關(guān)性,其中rs429358T/C位點突變基因頻率分布有統(tǒng)計學(xué)意義,提示該位點為新疆哈薩克族阿爾茨海默病的危險因素。
[Abstract]:Objective: to study the relationship between TCM syndromes of Alzheimer's disease and polymorphism of ApoE gene in Kazak nationality of Xinjiang. Methods:. The AD patients in Kazak nationality and the large clinical database of neurodegenerative diseases in Xinjiang were diagnosed on the basis of AD epidemiological investigation completed in Altay region of Xinjiang from June 2010 to November 2016. There were 58 cases. In the control group, 79 cases were classified according to the criteria of TCM diagnosis and common syndromes, and the genotypes of ApoE were detected in each group. Results: according to this experiment, the proportion of ApoE 蔚 2 carriers in two groups of Kazak population in Xinjiang was significantly lower than that in AD patients. The proportion of ApoE 蔚 4 carriers increased significantly. The proportion of ApoE 蔚 3 carriers was the highest in the normal population, followed by ApoE 蔚 2. ApoE 蔚 4 was the lowest. There was significant difference in allelic frequency between the two groups. The distribution of ApoE genotype after TCM typing was found in the case group and the control group. The distribution of ApoE 蔚 3 / 蔚 4 genotype in kidney essence deficiency syndrome was higher than that in control group (P 0.05). Compared with other syndromes, there was also a significant difference in the frequency distribution of mutation gene at rs429358T / C locus (P 0.05). For other loci, rs769449 / rs7412rs405509) there is no statistical significance in the number of existing samples. Conclusion: through the study of this subject. It was found that there was a correlation between the TCM syndrome type of Alzheimer's disease and the polymorphism of ApoE gene in Xinjiang Kazak nationality, and the frequency distribution of rs429358T/C locus mutation gene was statistically significant. These results suggest that this locus is a risk factor for Alzheimer's disease in Kazak nationality in Xinjiang.
