【摘要】：本課題開發(fā)出一種即食泥鰍食品工藝,利用其副產(chǎn)物魚頭為原料提取純化多糖并進(jìn)行結(jié)構(gòu)分析。通過(guò)工藝試驗(yàn),確立即食泥鰍脫腥液最優(yōu)配方為食鹽0.8%,碳酸鈉0.3%,β-環(huán)糊精0.7%；調(diào)味液配方為鹽18g、食醋40g、料酒40g、糖75g、醬油25g、特制辣醬35g、水100g。開發(fā)出的泥鰍即食食品具有良好的營(yíng)養(yǎng)和風(fēng)味,各種理化和微生物指標(biāo)均達(dá)到國(guó)標(biāo)標(biāo)準(zhǔn)。多糖的功能性質(zhì)與其結(jié)構(gòu)息息相關(guān),通過(guò)分析多糖結(jié)構(gòu)即可推斷該多糖的功能性質(zhì)。目前學(xué)術(shù)界對(duì)植物多糖和真菌多糖的研究很多,對(duì)于動(dòng)物多糖的研究報(bào)道并不多見。通過(guò)檢測(cè)即食泥鰍的營(yíng)養(yǎng)指標(biāo)可知該食品蛋白質(zhì)含量很高,但脂肪含量很低；泥鰍的頭、黏液以及全身含有大量功能性多糖,這為泥鰍高附加值產(chǎn)品開發(fā)提供了方向。多糖的提取純化和結(jié)構(gòu)分析實(shí)驗(yàn)方法主要有：泥鰍魚頭烘干粉碎、去脂、水提醇沉、savage法和酶法除蛋白、流水透析、過(guò)葡聚糖凝膠柱等過(guò)程來(lái)提取純化泥鰍多糖,利用Design-Expert 8.0統(tǒng)計(jì)軟件利用響應(yīng)面法優(yōu)化多糖提取條件；苯酚-硫酸法和3,5-二硝基水楊酸法分別測(cè)定純化后的泥鰍多糖總糖含量和還原糖含量；紫外光譜分析多糖純度；紅外光譜和核磁共振分析多糖官能團(tuán)結(jié)構(gòu)；高效液相色譜法測(cè)定多糖分子量；剛果紅實(shí)驗(yàn)驗(yàn)證多糖的空間結(jié)構(gòu)等。本課題實(shí)驗(yàn)結(jié)論如下：以粗多糖得率為響應(yīng)指標(biāo),利用design-expert8.0軟件進(jìn)行二次多元回歸擬合。所得公式如下：Y= 0.22+0.05X_1+0.22X_2+0.03X_3+0.063X_1X_2-0.2X_1X_3+0.027X_2X_3-0.19X_1~2 +6.75 × 10~(-3)X_2~2-0.15X_3~2由軟件分析得到泥鰍副產(chǎn)物多糖最優(yōu)提取工藝為：NaOH 0.11mol/L；乙醇濃度80%；浸提時(shí)間10h；粗多糖理論最高得率2.46%。同樣條件進(jìn)行驗(yàn)證試驗(yàn),得率為2.41%,與理論值基本相符,利用此方法對(duì)多糖提取條件優(yōu)化是可行的；總糖含量為79.51%,還原糖含量為0.27%,多糖含量為79.24%；紫外光譜表明多糖中無(wú)蛋白質(zhì)和核酸雜質(zhì),紅外光譜和核磁共振光譜圖表明多糖由β-構(gòu)型和α-構(gòu)型兩種吡喃糖組成且不含糖醛酸；高效液相色譜法測(cè)得多糖分子量7.11×105Da；剛果紅實(shí)驗(yàn)表明多糖有三股螺旋結(jié)構(gòu)。
[Abstract]:In this paper, a kind of instant food technology of Misgurnus anguillicaudatus was developed. The polysaccharide was extracted from the fish head, the by-product was used as the raw material, and the structure was analyzed. The optimum formula for deodorization of Loach was determined as follows: salt 0.8, sodium carbonate 0.33, 尾 -cyclodextrin 0.7.The seasoning solution formula was 18g salt, 40g vinegar, 40g cooking wine, 75g sugar, 25g soy sauce, 35g special chili sauce, 100g water. The instant food of Loach developed had good nutrition and flavor. The functional properties of polysaccharides are closely related to their structures. The functional properties of polysaccharides can be inferred by analyzing their structures. At present, there are a lot of studies on plant polysaccharides and fungal polysaccharides in academic circles. There are few reports of animal polysaccharides. The nutritional index of the instant Loach shows that the protein content of the food is high, but the fat content is very low, the head, mucus and body of Loach contain a lot of functional polysaccharides. The main methods of extraction, purification and structure analysis were as follows: drying and crushing, degreasing, water extraction and alcohol precipitation, enzymatic deproteinization, and income dialysis. The extraction and purification of Loach polysaccharides were carried out by using glucan gel column. The extraction conditions of the polysaccharides were optimized by using the response surface method (RSM) with Design-Expert 8.0 software. The content of total sugar and reducing sugar of purified Misgurnus anguillicaudatus polysaccharides were determined by phenol-sulfuric acid method and 3-dinitrosalicylic acid method respectively; the purity of polysaccharides was analyzed by ultraviolet spectrum; the structure of functional groups of polysaccharides was analyzed by IR and NMR. The molecular weight of polysaccharides was determined by HPLC, the spatial structure of polysaccharides was verified by Congo red experiment, and the results were as follows: the yield of crude polysaccharides was taken as the response index. The quadratic regression fitting was carried out by using design-expert8.0 software. The obtained formula was as follows: y = 0.22 0.05X_1 0.22X_2 0.03X_3 0.063X_1X_2-0.2X_1X_3 0.027X_2X_3-0.19X_1~2 6.75 脳 10~(-3)X_2~2-0.15X_3~2. The optimum extraction process of Misgurnus anguillicaudatus by-product polysaccharides was obtained by software analysis:: NaOH 0.11 mol / L; ethanol concentration 80%; extraction time 10 h; crude polysaccharide theory. The maximum yield is 2.46. The verification test is carried out under the same conditions, The yield is 2.41, which is basically consistent with the theoretical value. It is feasible to optimize the extraction conditions of polysaccharides by this method. The total sugar content is 79.51, the reducing sugar content is 0.27, and the polysaccharide content is 79.240.The ultraviolet spectrum shows that there are no protein and nucleic acid impurities in the polysaccharides. The IR and NMR spectra showed that the polysaccharides were composed of 尾 -configuration and 偽 -configuration and did not contain uronic acid, the molecular weight of polysaccharides was 7.11 脳 10 ~ 5Daby HPLC, and the Congo red experiment showed that the polysaccharides had three helical structures.
