固定化木聚糖酶的方法及酶學(xué)性質(zhì)的研究

【摘要】:本研究分別以殼聚糖、海藻酸鈉和D280大孔樹脂為載體制備固定化木聚糖酶,對其固定化條件進(jìn)行優(yōu)化,并對固定化酶的酶學(xué)性質(zhì)進(jìn)行了研究,得到以下結(jié)論:1)制備了殼聚糖固定化木聚糖酶,單因素試驗確定的最佳制備條件為:給酶量450U,固定化pH5.8,處理時間為5h,固定化溫度為40℃;正交試驗確定殼聚糖固定木聚糖酶的最優(yōu)條件為:給酶量450U,固定化pH5.6,處理時間為5h,固定化溫度為40℃,此時固定化木聚糖酶酶活力回收率最高,為35.6%;(2)制備了海藻酸鈉固定化木聚糖酶,單因素試驗確定的最佳制備條件為:給酶量360U,固定化pH5.8,處理時間為3h,固定化溫度為30℃;對固定化條件進(jìn)行正交試驗優(yōu)化,確定海藻酸鈉固定木聚糖酶的最優(yōu)條件為:給酶量360U,固定化pH5.6,處理時間為3h,處理溫度為30℃,此時固定化木聚糖酶酶活力回收率為39.3%;(3)制備了D280大孔樹脂固定化木聚糖酶,單因素試驗確定的最佳制備條件為:給酶量450U,固定化pH5.3,處理時間為7h,固定化溫度為50℃;對固定化條件進(jìn)行正交試驗優(yōu)化,確定D280大孔樹脂固定木聚糖酶的最優(yōu)條件為:給酶量450U,固定化pH5.1,處理時間為8h,固定化溫度為52℃,此時固定化木聚糖酶酶活力回收率為40.8%;(4)對三種載體制備的固定化酶酶學(xué)性質(zhì)進(jìn)行研究,與游離酶相比,三種固定化酶的儲藏穩(wěn)定性、熱穩(wěn)定性和酸堿穩(wěn)定性均優(yōu)于游離酶,其中D280大孔樹脂效果最好。(5)對三種載體制備的固定化酶的酶促反應(yīng)動力學(xué)進(jìn)行了研究,與游離酶相比,三種固定化酶的Km均有所升高,Vmax均有所降低,但D280大孔樹脂與游離酶相差最小,且基本處于同一水平,說明D280大孔樹脂制備的固定化酶與底物親和力最強。綜上所述,以D280大孔樹脂為載體固定化木聚糖酶在各項指標(biāo)上均優(yōu)于殼聚糖和海藻酸鈉。
[Abstract]:In this study, chitosan, sodium alginate and D280 macroporous resin were used as carriers to prepare immobilized xylanase, and the immobilized conditions were optimized, and the enzymatic properties of immobilized xylanase were studied.
