【摘要】:目的:在麻醉大鼠,觀察刺激視上核(supraoptic nucleus,SON)前后,延髓頭端腹外側(cè)區(qū)(rostral ventrolateral medulla,RVLM)神經(jīng)元單位放電以及外周生理指標的變化,探討SON和RVLM神經(jīng)元之間是否存在直接或間接的功能聯(lián)系,再進一步分析SON與RVLM間血管升壓素(vasopressin,VP)聯(lián)系的可能性,為闡明心血管功能的中樞調(diào)節(jié)機制提供新的理論依據(jù)。方法:取250~300g健康成年雄性SD大鼠,腹腔注射氨基甲酸乙酯(700mg/kg)和α-氯醛糖(35 mg/kg)混合麻藥麻醉后,固定于腦立體定位儀,采用Powerlab多通道記錄系統(tǒng)以及A-M Systems的刺激器和放大器系統(tǒng),進行大鼠RVLM神經(jīng)元單位放電、股動脈血壓、心電圖、呼吸肌肌電以及體溫的同步記錄。然后將金屬刺激電極置于SON核團處,觀察電刺激SON前后RVLM神經(jīng)元單位放電以及其它外周指標的變化,分析SON與RVLM間是否存在功能聯(lián)系。在RVLM核團微量注射VP受體(V1a受體)阻斷劑SR49059,觀察注射SR49059前后,電刺激SON,RVLM神經(jīng)元單位放電及外周指標的改變,以進一步分析SON與RVLM間VP聯(lián)系的可能性。結(jié)果:1.心血管中樞與心血管功能的同步記錄技術(shù):在18只大鼠共記錄到23個RVLM神經(jīng)元自發(fā)單位放電,其中有16個是連續(xù)性放電型式,7個是周期性簇狀放電型式。兩種類型放電分別對應(yīng)的同步記錄指標進行組間比較,RVLM放電頻率、RVLM放電幅值、心率變異性(heart rate variability,HRV)(LF/HF)、心率、體溫、收縮壓、舒張壓、平均動脈壓以及呼吸肌肌電頻率均無統(tǒng)計學(xué)差異,僅呼吸肌肌電幅值變化有統(tǒng)計學(xué)意義(P0.05)。血壓升高時,觀察到RVLM神經(jīng)元單位放電明顯被抑制;針刺大鼠尾部引起RVLM神經(jīng)元放電活動出現(xiàn)變化,從而使外周各指標也發(fā)生相應(yīng)改變。2.電刺激SON對RVLM神經(jīng)元單位放電及外周指標的影響:在18只SD大鼠共記錄到19個RVLM神經(jīng)元單位放電。刺激(200μA,0.2ms,100Hz,0.2s)SON后,觀察到10個RVLM神經(jīng)元單位放電頻率顯著下降(P0.05),放電幅值下降,相應(yīng)的平均動脈壓上升(P0.05),心率、呼吸頻率均有下降趨勢;9個神經(jīng)元單位放電頻率顯著升高(P0.05),其它指標變化不顯著。3.RVLM核團微量注射SR49059的作用:在RVLM注射SR49059預(yù)處理前,對SON的200μA電刺激在9個RVLM神經(jīng)元觀察到單位放電頻率顯著下降(P0.05),單位放電幅值下降,心率呈下降趨勢。在SR49059預(yù)處理后,觀察到RVLM神經(jīng)元單位放電頻率和幅值均呈上升趨勢,其它外周指標分析均無統(tǒng)計學(xué)差異;在注入SR49059立即電刺激SON,觀察到各項指標無顯著變化;在注入SR49059穩(wěn)定10min后電刺激SON,觀察到RVLM神經(jīng)元單位放電頻率和幅值無明顯下降,其他指標亦未見顯著變化。對SON進行400μA電刺激后,也觀察到RVLM神經(jīng)元單位放電頻率和幅值呈下降趨勢;心率、LF/HF呈下降趨勢。同樣,RVLM注射SR49059預(yù)處理后,RVLM神經(jīng)元單位放電頻率和幅值均呈上升趨勢,其它外周指標分析均無統(tǒng)計學(xué)差異;在注入SR49059后立即電刺激SON,各項指標分析均無統(tǒng)計學(xué)差異;在注入SR49059穩(wěn)定10min后電刺激SON,RVLM神經(jīng)元單位放電頻率和幅值呈下降趨勢,其他指標未見顯著變化。結(jié)論:(1)所建立的心血管中樞神經(jīng)元單位放電和外周生理指標的同步記錄技術(shù),適用于觀察心血管各級中樞之間的相互作用,以及心血管中樞與心血管外周活動之間的相互影響。(2)電刺激SON,對RVLM不同心血管相關(guān)神經(jīng)元作用不同,表現(xiàn)為抑制或興奮作用,并伴有外周指標的相應(yīng)改變,提示SON與RVLM間存在直接或間接功能聯(lián)系的可能性,且對心血管活動有一定的調(diào)節(jié)作用。(3)RVLM核團微量注射VP受體阻斷劑SR49059后,電刺激SON后RVLM神經(jīng)元放電活動抑制作用減弱,提示VP可能參與該通路功能聯(lián)系。
[Abstract]:Objective: in anesthetized rats, observe the stimulation of supraoptic nucleus (supraoptic, nucleus, SON) and the rostral ventrolateral medulla (rostral ventrolateral, medulla, RVLM) change the unit discharges of neurons and peripheral physiological indexes, discuss whether there is direct or indirect links between SON and RVLM neurons, and further analysis of SON RVLM vasopressin (vasopressin, VP) with the possibility of providing a new theoretical basis for clarifying central cardiovascular function regulation mechanism. Methods: 250~300g healthy adult male SD rats by intraperitoneal injection of urethane (700mg/kg) and alpha chloralose (35 mg/kg) mixed anesthesia after anesthesia, fixed in the brain stereotaxic apparatus, using Powerlab multi channel recording system and A-M Systems stimulator and amplifier system of neurons in rat RVLM, femoral arterial blood pressure, ECG, respiratory muscle and body temperature The synchronous record. Comparison of two types of discharge index recorded respectively between the groups, the discharge frequency of RVLM, RVLM discharge amplitude, heart rate variability (heart rate, variability, HRV) (LF/HF), Heart rate, body temperature, systolic blood pressure, diastolic blood pressure, there was no significant difference in mean arterial pressure and respiratory muscle EMG frequency, only respiratory muscle EMG amplitude change was statistically significant (P0.05). Blood pressure, observed the unit discharge of RVLM neurons were inhibited obviously by acupuncture; rat tail caused changes in firing of RVLM neurons, which the peripheral indicators of the impact on the change of.2. SON stimulation on unit discharges of RVLM neurons and peripheral index: 18 SD rats were recorded to discharge 19 RVLM neurons. Stimulation (200 A, 0.2ms, 100Hz, SON, 0.2S) were significantly decreased to the discharge rate of 10 RVLM neurons (P0.05), discharge amplitude decreased, the mean arterial pressure increased accordingly (P0.05), heart rate, respiratory rate decreased; the discharge frequency of 9 units increased significantly (P0.05), the other indexes did not change significantly.3.RVLM nucleus trace The role of SR49059 in RVLM injection: the injection of SR49059 before the pretreatment of 200 A electric stimulation, SON to the unit discharge were observed in 9 neurons in the RVLM frequency decreased significantly (P0.05), the unit discharge amplitude decreased, heart rate decreased in SR49059. After the pretreatment, the observed frequency and amplitude of unit discharge of RVLM neurons were the rise, other peripheral indexes were not statistically significant; the SR49059 injection in the electrical stimulation of the SON immediately, observed no significant changes in the indicators; SR49059 injection in the SON stable 10min after electrical stimulation, unit discharge of RVLM neurons observed frequency and amplitude decreased significantly, there was no significant changes in other indicators of 400 A. Electrical stimulation of the SON, also observed the frequency and amplitude of unit discharge of RVLM neurons was decreased; heart rate, LF/HF decreased. Similarly, RVLM injection after pretreatment with SR49059, a single RVLM neuron discharge frequency and amplitude Showed an upward trend, other peripheral index analysis showed no significant differences; in the injection of SR49059 immediately after SON stimulation, there was no significant difference in each index analysis; SR49059 SON injection stimulation stable after 10min, the frequency and amplitude of unit discharge of RVLM neurons decreased, other indexes had no significant changes. Conclusion: (1) the technique of simultaneous recording discharge and peripheral physiological indexes of neurons cardiovascular center established, the interaction between observation for cardiovascular center at all levels, the mutual influence between central and peripheral cardiovascular and cardiovascular activities. (2) the electrical stimulation of SON, different RVLM of different cardiovascular related neurons showed inhibitory or excitatory effect. The corresponding change, accompanied by peripheral index, suggesting the existence of possibility of direct or indirect connections between RVLM and SON function, and has a certain role in the regulation of cardiovascular activity. (3) RV After microinjection of VP receptor blocker SR49059 in the LM nucleus, the inhibitory effect of RVLM neuron discharge activity was weakened after the electrical stimulation of SON, suggesting that VP may be involved in the function of the pathway.
