異養(yǎng)硝化菌Acinetobacter sp.的分離鑒定及其脫氮特性

【摘要】:從北京市高碑店污水處理廠(chǎng)BBR(Bacillus Bacteria Bioreactor)中試設(shè)備的活性污泥樣品中分離得到一株異養(yǎng)硝化-好氧反硝化菌株JQ1004,利用16S r DNA對(duì)其進(jìn)行同源性分析,初步鑒定其為不動(dòng)桿菌屬(Acinetobacter sp.).在好氧條件下,以丁二酸鈉為碳源,分別以氨氮和硝酸鹽氮為氮源,研究其脫氮特性,發(fā)現(xiàn)該菌株在33h時(shí)氨氮去除率達(dá)到99.45%,COD去除率達(dá)到92.54%;在34h時(shí),硝酸鹽氮去除率達(dá)到84.42%,而COD去除率達(dá)到93.11%.另外通過(guò)研究發(fā)現(xiàn),與降解氨氮相比,菌株JQ1004在降解硝酸鹽氮時(shí)需要更高的C/N.利用Compertz模型對(duì)其脫氮特性進(jìn)行擬合發(fā)現(xiàn),菌株對(duì)氨氮的最大降解速率明顯高于降解硝酸鹽氮的最大降解速率,分別為Rm=7.93mg/(L·h)和Rm=4.08mg/(L·h).利用響應(yīng)面法進(jìn)行實(shí)驗(yàn)設(shè)計(jì),優(yōu)化其脫氮條件,得到最佳脫氮條件為:pH=7.33;溫度=31.80℃;轉(zhuǎn)速=154.54r/min;C/N=7.76.
[Abstract]:A heterotrophic Nitrification-aerobic denitrification strain JQ1004 was isolated from the activated sludge samples of BBR (Bacillus Bacteria Bioreactor) pilot plant of Beijing Gaobeidian Sewage Treatment Plant. The strain was identified as Acinetobacter sp. Under aerobic conditions, sodium succinate was used as the catalyst. Carbon source, using ammonia nitrogen and nitrate nitrogen as nitrogen sources, the denitrification characteristics of the strain were studied. It was found that the removal rate of ammonia nitrogen and COD reached 99.45% and 92.54% respectively at 33H and 84.42% and 93.11% respectively at 34h. A higher C/N ratio is needed. The results show that the maximum degradation rate of ammonia nitrogen is obviously higher than that of nitrate nitrogen, Rm=7.93 mg/(L.h) and Rm=4.08 mg/(L.h). Response surface methodology is used to optimize the denitrification conditions. PH=7.33; temperature =31.80 C; speed =154.54r/min; C/N=7.76.
