抗生素抗性基因在養(yǎng)殖廢水中的分布與去除

【摘要】:畜禽養(yǎng)殖場(chǎng)作為抗生素抗性基因的一個(gè)熱點(diǎn)區(qū),其產(chǎn)生的廢水中存在大量的抗生素抗性基因,直接排放將會(huì)污染接受的水體.本研究以微生物固化曝氣技術(shù)為核心,構(gòu)建了一個(gè)包含沉淀池、一級(jí)處理池、二級(jí)處理池和氧化塘的養(yǎng)殖廢水處理系統(tǒng).采用高通量熒光定量PCR技術(shù),探究各種抗性基因在進(jìn)水、各處理池和出水中的種類、多樣性和豐度的變化以及可能的影響因素.研究發(fā)現(xiàn):(1)微生物固化曝氣技術(shù)能有效降低養(yǎng)殖廢水中抗性基因的種類和多樣性;(2)該技術(shù)也能有效去除養(yǎng)殖廢水中抗性基因的絕對(duì)豐度,去除率高達(dá)93.6%;(3)養(yǎng)殖廢水出水中抗性基因絕對(duì)豐度仍高于自然水體,其直接排放仍有抗性基因污染風(fēng)險(xiǎn);(4)畜禽養(yǎng)殖廢水中TN和土霉素的含量與許多抗生素抗性基因總豐度存在正相關(guān)性.通過(guò)對(duì)TN和土霉素的去除或者控制使用,可以有效降低養(yǎng)殖廢水中的抗性基因.
[Abstract]:As a hot spot of antibiotic resistance genes, livestock farms produce a large number of antibiotic resistance genes in their wastewater, which will pollute the accepted water body directly. Based on microbial solidified aeration technology, a system of aquiculture wastewater treatment including sedimentation tank, primary treatment tank, secondary treatment tank and oxidation pond was constructed. High throughput fluorescence quantitative PCR technique was used to investigate the variety, diversity and abundance of various resistance genes in the influent, in each treatment tank and in the effluent, as well as the possible influencing factors. It is found that the microorganism solidified aeration technology can effectively reduce the species and diversity of resistance genes in aquaculture wastewater. This technique can also effectively remove the absolute abundance of resistance genes in aquaculture wastewater. The absolute abundance of resistance gene in the effluent of culture wastewater is still higher than that in the natural water. The content of TN and oxytetracycline in livestock and poultry wastewater was positively correlated with the total abundance of many antibiotic resistance genes. Removal or control of TN and oxytetracycline can effectively reduce resistance genes in culture wastewater.
