污泥中嗜酸酵母菌的分離鑒定及其重金屬耐受性研究

【摘要】:從污水處理廠的活性污泥中分離純化出一株嗜酸異養(yǎng)菌,命名為JJU-2,并對其進(jìn)行了形態(tài)觀察、生理生化特性測試及26SrDNA的D1/D2區(qū)序列分析,鑒定該菌株為假絲酵母菌(Candida palmioleophila)。進(jìn)一步對JJU-2菌株的生長特性和重金屬耐受性進(jìn)行研究。結(jié)果表明:該菌株最適生長pH為3~6,且能適應(yīng)pH3的極端酸性環(huán)境,最適生長溫度為25℃,最高耐受溫度為40℃,當(dāng)培養(yǎng)至3h,JJU-2菌株進(jìn)入對數(shù)生長期,培養(yǎng)至18h進(jìn)入穩(wěn)定期。Cr~(3+)、Ni~(2+)、Pb~(2+)對JJU-2菌株的最低抑菌摩爾濃度分別為20、2.0、40mmol/L。
[Abstract]:An acidophilic heterotrophic strain named JJU-2 was isolated and purified from activated sludge of wastewater treatment plant. It was identified as Candida palmiophilophilia by morphological observation, physiological and biochemical characteristics test and D1/D2 sequence analysis of 26SrDNA. The growth characteristics and heavy metal tolerance of JJU-2 strain were further studied. The results showed that the optimum growth pH of the strain was 3 ~ 6, and it could adapt to the extremely acidic environment of pH3. The optimum growth temperature was 25 鈩,
