釀酒廢水產(chǎn)微生物絮凝劑菌株的篩選及發(fā)酵條件優(yōu)化

【摘要】:該研究采用平板劃線法、液體發(fā)酵篩選獲得產(chǎn)絮凝劑菌株,命名為Y1,采用形態(tài)學(xué)觀察、16S r DNA測(cè)序及系統(tǒng)進(jìn)化樹分析進(jìn)行鑒定;并在單因素試驗(yàn)基礎(chǔ)上,采用響應(yīng)面試驗(yàn)設(shè)計(jì)方法優(yōu)化菌株Y1的發(fā)酵條件。結(jié)果表明,菌株Y1為枯草芽孢桿菌(Bacillus subtilis),最佳發(fā)酵條件為初始p H 7.4,轉(zhuǎn)速147 r/min,溫度35℃,接種量8%。在最佳發(fā)酵條件下,絮凝率為81.6%。
[Abstract]:In this study, a flocculant producing strain, named Y1, was obtained by liquid fermentation, and was identified by morphological observation, 16s r DNA sequencing and phylogenetic tree analysis. The fermentation conditions of strain Y1 were optimized by response surface design. The results showed that strain Y1 was Bacillus subtilisa.
