釀酒廢水產(chǎn)微生物絮凝劑菌株的篩選及發(fā)酵條件優(yōu)化

【摘要】:該研究采用平板劃線法、液體發(fā)酵篩選獲得產(chǎn)絮凝劑菌株,命名為Y1,采用形態(tài)學(xué)觀察、16S r DNA測(cè)序及系統(tǒng)進(jìn)化樹分析進(jìn)行鑒定;并在單因素試驗(yàn)基礎(chǔ)上,采用響應(yīng)面試驗(yàn)設(shè)計(jì)方法優(yōu)化菌株Y1的發(fā)酵條件。結(jié)果表明,菌株Y1為枯草芽孢桿菌(Bacillus subtilis),最佳發(fā)酵條件為初始p H 7.4,轉(zhuǎn)速147 r/min,溫度35℃,接種量8%。在最佳發(fā)酵條件下,絮凝率為81.6%。
[Abstract]:In this study, a flocculant producing strain, named Y1, was obtained by liquid fermentation, and was identified by morphological observation, 16s r DNA sequencing and phylogenetic tree analysis. The fermentation conditions of strain Y1 were optimized by response surface design. The results showed that strain Y1 was Bacillus subtilisa. The optimum fermentation conditions were as follows: initial pH 7.4, rotational speed 147 rmin, temperature 35 鈩,
