雙塞頭固定化細胞反應器降解TCE

【摘要】:為探究固定化細胞對三氯乙烯(TCE)的生物降解特性及機理,以優(yōu)選的活性炭纖維為固定化材料,構建雙塞頭固定化細胞反應器;利用填埋場覆蓋層富集篩選的高效混合菌SWA1為生物介質開展細胞固定化及TCE生物降解研究;運用掃描電鏡,熒光定量PCR和高通量測序技術分別考察TCE降解前后固定化微生物微觀結構、關鍵酶表達豐度和混合菌群落結構變化.結果顯示:(1)該固定化細胞反應器對微生物具有良好的固定化效果,且微生物可維持較高活性(甲烷氧化速率為0.6μmol g~(-1) d~(-1));(2)在TCE初始濃度為18 mg/L時,反應1 d降解率為94.1%,降解過程中有8.7μmol有機氯轉化為氯離子;(3)反應器中TCE降解關鍵酶為甲烷單加氧酶和苯酚羥化酶,TCE降解后優(yōu)勢菌屬為Anaerolineaceae_unclassified(0.6%-19.9%)和甲基孢囊菌屬Methylocystis(0-15.8%).該雙塞頭固定化細胞反應器能高效去除氯代烴類污染物,可為含氯有機廢水高效生物治理的工程應用提供參考.
[Abstract]:In order to investigate the biodegradation characteristics and mechanism of TCEs by immobilized cells, a double-plug immobilized cell reactor was constructed using the selected activated carbon fiber as the immobilized material. Cell immobilization and biodegradation of TCE were carried out by using highly efficient mixed bacteria (SWA1) enriched and screened in landfill as biomaterials. The microstructures of immobilized microorganisms before and after TCE degradation were investigated by scanning electron microscopy fluorescence quantitative PCR and high-throughput sequencing. The results showed that the immobilized cell reactor had a good immobilization effect on microorganism. The microorganism can maintain high activity (methane oxidation rate is 0. 6 渭 mol / g ~ (-1)). (2) when the initial concentration of TCE was 18 mg/L, the degradation rate for 1 day was 94.1, and there was 8.7 渭 mol organic chlorine converted to chloride ion in the degradation process. The key enzymes of TCE degradation in the reactor are methane monooxygenase and phenol hydroxylase. The dominant bacteria of the genus Anaerolineaceaeae after TCE degradation are 0.6-19.9- 0.6-19.9in the genus Anaerolineaceaeae. And Methylocystis 0-15.80.The double plug immobilized cell reactor can effectively remove chlorinated hydrocarbon pollutants. It can provide reference for the engineering application of high efficiency biological treatment of organic wastewater containing chlorine.
