枸杞多糖對(duì)腦力疲勞型心理亞健康模型大鼠認(rèn)知功能的影響

【摘要】:目的:建立腦力疲勞型心理亞健康大鼠模型,并觀察枸杞多糖對(duì)腦力疲勞型心理亞健康大鼠認(rèn)知功能的影響,為心理亞健康的預(yù)防與干預(yù)提供理論指導(dǎo)。方法:取清潔級(jí)Sprague Dawley大鼠50只,按照體重分層隨機(jī)分為5組,即正常空白對(duì)照組(C,n=10),模型空白對(duì)照組(PS,n=10),模型給生理鹽水組(PS+NS,n=10),模型給枸杞多糖低劑量組(PS+LBP20mg/kg,n=10),模型給枸杞多糖高劑量組(PS+LBP40mg/kg,n=10)。造模程序分為環(huán)境適應(yīng)期、空瓶不確定應(yīng)激飲水訓(xùn)練期和正式造模三個(gè)階段。采用復(fù)合應(yīng)激因素進(jìn)行模型的構(gòu)建,模擬現(xiàn)代社會(huì)腦力工作者長(zhǎng)期睡眠不足、久坐不動(dòng)活動(dòng)量較少、飲食不規(guī)律和工作當(dāng)中不確定出現(xiàn)的應(yīng)激壓力以及人際交往問題來進(jìn)行模型的構(gòu)建。從正式造模的第1天起至造模結(jié)束的第21天,采用可變計(jì)量法分別給予PS+NS組、PS+LBP20mg/kg組和PS+LBP40mg/kg組大鼠相應(yīng)劑量的灌胃給藥。造模結(jié)束后,通過鑒別大鼠是否處于疾病狀態(tài)、進(jìn)行活動(dòng)性與情緒的檢驗(yàn)和學(xué)習(xí)記憶實(shí)驗(yàn)來檢驗(yàn)?zāi)X力疲勞型心理亞健康的主要核心癥狀,對(duì)新模型和藥物的干預(yù)進(jìn)行檢驗(yàn)和評(píng)價(jià)。結(jié)果:1.腦力疲勞型心理亞健康大鼠模型的鑒定C組大鼠與PS組大鼠第7天、第14天、第21天的體重以及體重增長(zhǎng)量差異均具有統(tǒng)計(jì)學(xué)意義(P0.01)。PS組大鼠體重低于C組大鼠,且體重相比C組明顯增長(zhǎng)緩慢。C組與PS組兩組大鼠的曠場(chǎng)垂直站立次數(shù)、曠場(chǎng)總距離和平均速度差異顯著(P0.05)。兩組大鼠的曠場(chǎng)水平運(yùn)動(dòng)次數(shù)差異顯著(P0.01)。C組與PS組兩組大鼠的開放臂滯留時(shí)間比例方面的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。C組與PS組兩組大鼠在Day6、Day7八臂迷宮的測(cè)試完成時(shí)間差異具有統(tǒng)計(jì)學(xué)意義(P0.05)。C組與PS組兩組大鼠在Day6八臂迷宮的工作記憶錯(cuò)誤次數(shù)具有統(tǒng)計(jì)學(xué)意義(P0.05)。2.枸杞多糖干預(yù)對(duì)五組大鼠的影響(1)C組21天的體重與PS+LBP40mg/kg組、PS+LBP20mg/kg組PS+NS組和PS組組間比較均具有統(tǒng)計(jì)學(xué)意義(P0.01)。PS+LBP40mg/kg組、PS+LBP20mg/kg組21天的體重與PS+NS組和PS組組間比較均具有統(tǒng)計(jì)學(xué)意義(P0.01)。第1周、第2周、第3周的體質(zhì)量增長(zhǎng)量各組大鼠的組間差異均具有統(tǒng)計(jì)學(xué)意義(P0.01)。(2)C組與PS組的曠場(chǎng)總距離、平均速度、垂直站立次數(shù)、水平跨格數(shù)比較差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。PS+LBP20mg/kg組與PS組的曠場(chǎng)總距離、垂直站立次數(shù)、曠場(chǎng)水平跨格數(shù)比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。PS+LBP40mg/kg組與PS組的曠場(chǎng)垂直站立次數(shù)、水平跨格數(shù)比較差異有統(tǒng)計(jì)學(xué)意義(P0.05),(3)C組與PS+NS組開放臂進(jìn)入次數(shù)比例、開放臂滯留時(shí)間比例均具有統(tǒng)計(jì)學(xué)意義(P0.05),PS組與C組、PS+LBP40mg/kg組開放臂進(jìn)入次數(shù)比例差異有統(tǒng)計(jì)學(xué)意義(P0.05)。PS組與C組、PS+LBP40mg/kg組、PS+LBP20mg/kg組開放臂滯留時(shí)間比例差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(4)八臂迷宮測(cè)試第3天PS+LBP40mg/kg與PS+NS組的測(cè)試時(shí)間差異有統(tǒng)計(jì)學(xué)意義(P0.05)。八臂迷宮測(cè)試第6天C組與PS+NS組、PS組參考記憶錯(cuò)誤次數(shù)比較(P0.05);PS+LBP40mg/kg與PS參考記憶錯(cuò)誤次數(shù)比較(P0.05);PS+LBP20mg/kg組與PS+NS參考記憶錯(cuò)誤次數(shù)比較(P0.05)。八臂迷宮測(cè)試第7天C組與PS+NS組、PS組參考記憶錯(cuò)誤次數(shù)比較(P0.05)。八臂迷宮測(cè)試第6天C組與PS+NS組、PS組正確率比較(P0.05),PS+LBP40mg/kg與PS組正確率比較(P0.05)。結(jié)論:1.通過慢性不完全睡眠剝奪、活動(dòng)限制、飲食不規(guī)律、空瓶不確定性心理應(yīng)激和社交失敗復(fù)合應(yīng)激因素進(jìn)行21天的腦力疲勞型心理亞健康大鼠模型的構(gòu)建,從一般情況觀察、體溫、體重、曠場(chǎng)實(shí)驗(yàn)、高架十字迷宮實(shí)驗(yàn)和八臂迷宮實(shí)驗(yàn)檢測(cè)指標(biāo)的分析結(jié)果認(rèn)為,腦力疲勞型心理亞健康模型的構(gòu)建基本成功。2.腦力疲勞型心理亞健康模型大鼠的癥狀表現(xiàn)主要為體重增長(zhǎng)緩慢、精神緊張或煩躁不安造成的自主活動(dòng)性活動(dòng)增加、焦慮的情緒體驗(yàn)以及學(xué)習(xí)記憶能力的下降等。3.LBP在模型構(gòu)建的過程中對(duì)腦力疲勞型心理亞健康模型大鼠的一般情況、自主活動(dòng)情況、焦慮緊張情緒以及學(xué)習(xí)記憶下降等問題具有一定程度的預(yù)防和改善作用。
[Abstract]:Objective: to establish a mental fatigue model of Mental Subhealth, and to observe the effect of Lycium barbarum polysaccharides on cognitive function of mental fatigue Mental Subhealth rats, and to provide theoretical guidance for the prevention and intervention of Mental Subhealth. Methods: 50 clean Sprague Dawley rats were selected and divided into 5 groups according to the weight stratification, that is, normal blank control group (C N=10), the model blank control group (PS, n=10), the model to the saline group (PS+NS, n=10), the model to the low dose group of Lycium barbarum polysaccharide (PS+LBP20mg/kg, n=10), the model to the high dose group (PS+LBP40mg/kg, n=10) of Lycium barbarum polysaccharide (PS+LBP40mg/kg, n=10). The modeling program is divided into the environmental adaptation period, the empty bottle uncertainty stress drinking training period and the formal model three stages. The model was constructed to simulate the lack of long-term sleep, less sedentary activity, irregular diet, stress stress and interpersonal problems in the modern society. From the first day of the formal model to the twenty-first day of the end of the model, the variable measurement method was adopted. The PS+NS group, PS+LBP20mg/kg group and group PS+LBP40mg/kg rats were given the corresponding dose of gavage.
