內(nèi)源乳化法制備溶菌酶腸溶性微膠囊及所用油相回收的研究

【摘要】:溶菌酶是一種作用于微生物細胞壁的抗菌蛋白,因其較強的抗菌作用被廣泛應用于飼料行業(yè),食品行業(yè)和醫(yī)藥行業(yè)等領域。但在實際應用過程中,溶菌酶作為腸溶性抗菌藥物時,因消化道胃酸的影響,使溶菌酶到達腸內(nèi)時的抗菌性降低。本課題針對此問題,采用微膠囊技術(shù),選擇腸溶性壁材—殼聚糖和海藻酸鈉,用內(nèi)源乳化法制備溶菌酶腸溶性微膠囊。同時,除了用石蠟油作為油相,還研究了安全性高的可食用菜籽油作為乳化油相制備微膠囊,使該工藝制備的產(chǎn)品更好的應用于食品、醫(yī)藥等行業(yè)。最后,為了使得該制備工藝具有更好的合理性和實際性,本課題首次研究重復利用工藝中乳化的油相,并重復利用,取得了比較理想的效果。主要研究結(jié)果如下: 首先采用內(nèi)源乳化法制備溶菌酶微膠囊。最佳配比:海藻酸鈉濃度2.0%,溶菌酶濃度1.5%,殼聚糖濃度0.3%,Span80的添加量為油相體積的1%。正交試驗得出制備微膠囊最佳工藝條件:碳酸鈣與海藻酸鈉質(zhì)量比7:40,水相與油相體積比30:70,冰醋酸與碳酸鈣摩爾比為3:1。最佳制備條件下溶菌酶微膠囊包封率為89.2%,載藥量為34.9%;在人工模擬胃液中2h后的釋放率為35.5%,在人工模擬腸液中4h后的總釋放率為88.9%。在模擬胃液中2h后,包埋后的溶菌酶活性保留率為89.5%,未包埋的僅為31.7%,包埋后的溶菌酶活性保留率比未包埋時明顯提高。 然后,用石蠟油和菜籽油分別作為乳化油相制備溶菌酶微膠囊。檢測微膠囊性能結(jié)果顯示分別為:包封率為89.2%和81.7%,載藥量為34.9%和26.6%,胃內(nèi)釋放率為35.5%和38.7%,胃腸液中總釋放率為88.9%和85.4%,包埋的溶菌酶在胃液中2h后,活性保留率為89.5%和84.7%。用菜籽油制備的微膠囊效果低于用石蠟油制備的。 其次,分別研究了廢棄乳狀液中石蠟油和菜籽油的回收方法。在石蠟油回收中,比較了加熱法,無機鹽結(jié)合加熱法,離心后加熱法三種方法對石蠟油的回收效果,結(jié)果顯示離心后加熱法相對較好,在離心轉(zhuǎn)速10000rpm下,離心10min,然后60℃水浴5min,油回收率為98.2%。與石蠟油相比,菜籽油相對更容易進行回收,通過高速離心法,短時間內(nèi)即可完成對菜籽油的回收,條件為離心轉(zhuǎn)速為10000rpm,離心時間為10min。 最后,研究了石蠟油和菜籽油的重復利用。用回收的石蠟油和菜籽油制備微膠囊,所得到的微膠囊的性能明顯下降。
[Abstract]:The lysozyme is an antibacterial protein acting on the cell wall of the microorganism, and is widely used in the fields of feed industry, food industry and medicine industry because of its strong antibacterial effect. In the actual application, however, when the lysozyme is used as the enteric-soluble antibacterial agent, the antibacterial property of the lysozyme to the intestine is reduced due to the influence of the gastric acid in the digestive tract. In this paper, the micro-capsule technology was used to select the enteric-soluble wall material, chitosan and sodium alginate, and the enteric-soluble microcapsule was prepared by the internal emulsification method. At the same time, in addition to using paraffin oil as the oil phase, the edible rapeseed oil with high safety is also studied as the emulsified oil phase to prepare the microcapsule, so that the product prepared by the process is better applied to the industries of food, medicine and the like. Finally, in order to make the preparation process more reasonable and practical, the oil phase emulsified in the process is studied for the first time, and the application is repeated, and the ideal effect is obtained. The main results are as follows: in that first place, the lysozyme is prepare by using the endogenous emulsification method, The optimal ratio of the capsule is as follows: the concentration of the sodium alginate is 2.0%, the concentration of the lysozyme is 1.5%, the concentration of the chitosan is 0.3%, and the amount of the Span 80 is the volume of the oil phase. The optimum conditions for preparing the microcapsules were obtained by orthogonal test: the mass ratio of calcium carbonate to sodium alginate was 7:40, the volume ratio of the aqueous phase to the oil phase was 30:70, and the molar ratio of glacial acetic acid to calcium carbonate was 3. 1. The entrapment efficiency of the lysozyme was 89.2% and 34.9% in the best preparation condition. The release rate was 35.5% after 2 h in the artificial simulated gastric juice, and the total release rate after 4 h in the artificial simulated intestinal fluid was 88. After 2 h of the simulated gastric juice, the active retention of the embedded lysozyme was 89.5%, the non-embedded lysozyme activity was 31.7%, and the retention rate of the embedded lysozyme was higher than that of the non-embedded lysozyme. and then, the paraffin oil and the rapeseed oil are used as the emulsified oil phase to prepare the solution, The results showed that the encapsulation rate was 89.2% and 81.7%, the drug-loading rate was 34.9% and 26.6%, the intragastric release rate was 35.5% and 38.7%, the total release rate in the gastrointestinal fluid was 88.9% and 85.4%, and the retained lysozyme was 89.5% after 2 h of gastric juice. 84.7%.
