抑制線粒體復(fù)合體Ⅱ誘導(dǎo)線粒體自噬影響細胞增殖的研究(英文)

線粒體復(fù)合體Ⅱ,也被稱為琥珀酸脫氫酶,參與線粒體呼吸作用及代謝重編程的調(diào)控過程.復(fù)合體Ⅱ由4個亞基構(gòu)成,其突變與腫瘤的發(fā)生密切相關(guān).本文探討復(fù)合體Ⅱ與線粒體自噬調(diào)控及細胞增殖之間的關(guān)系.實驗采用復(fù)合體Ⅱ的特異性抑制劑TTFA或敲除復(fù)合體Ⅱ的B亞基(SDHB)使其功能缺失.結(jié)果發(fā)現(xiàn),復(fù)合體Ⅱ功能的缺失顯著引起線粒體形態(tài)的片段化進而發(fā)生線粒體自噬,導(dǎo)致線粒體蛋白水平減少,抑制ATP生成;由于線粒體功能受到抑制,細胞葡萄糖消耗及乳酸產(chǎn)生水平增加,并顯著抑制細胞的細胞的增殖.綜上所述,復(fù)合體Ⅱ功能缺失可能通過調(diào)控線粒體自噬而影響細胞增殖,從而在腫瘤發(fā)生中起重要作用.
