Trio偶聯(lián)膜轉(zhuǎn)運機制調(diào)節(jié)神經(jīng)元突起生長

神經(jīng)元突起的生長和導向調(diào)節(jié)是神經(jīng)發(fā)育生物學的基本問題,而細胞骨架重排和膜運輸是神經(jīng)突起生長和導向的基本過程,研究該過程的調(diào)節(jié)規(guī)律是神經(jīng)發(fā)育生物學的關鍵課題。大量研究表明,神經(jīng)細胞可通過多條信號通路分別調(diào)節(jié)細胞骨架重排和膜運輸過程,但這兩個過程是否存在偶聯(lián)調(diào)節(jié)機制目前還不清楚。本文利用含復雜功能域的Trio分子模型,系統(tǒng)研究了該分子的調(diào)節(jié)規(guī)律、并提出了一個細胞骨架重排和膜運輸?shù)呐悸?lián)機制。Trio分子包含兩個鳥苷酸交換因子結構域(GEFD)、一個未知功能的蛋白激酶結構、多個Spectrin結構域等,在體內(nèi)還存在有4個不同長度的剪切體。我們推測,這些結構的復雜性可能是神經(jīng)元生長精細調(diào)節(jié)或協(xié)同調(diào)節(jié)的物質(zhì)基礎。為研究Trio多功能域間的協(xié)調(diào)調(diào)節(jié)規(guī)律,我們試圖對不同結構域、不同剪切體進行系統(tǒng)地功能解析。我們將兩個鳥苷酸交換因子結構域GEF1和GEF2分別進行片段敲除,并成功地建立了可遺傳的小鼠模型(G1NKO和G2NKO)。遺憾的是,G1NKO小鼠完全不表達Trio蛋白,其表型與Trio全蛋白敲除小鼠完全相同,提示該片段敲除導致了全蛋白敲除,其機制目前還不知曉。有意思的是,GEFD2片段敲除小鼠也...
