稻瘟菌免疫誘導(dǎo)蛋白MoSM1互作蛋白的篩選及驗證

稻瘟菌分泌的小分子蛋白MoSM1是一類Cerato-platanin蛋白。目前,已經(jīng)發(fā)現(xiàn)MoSM1蛋白作為稻瘟菌的毒力因子也可以作為植物免疫反應(yīng)的激發(fā)子。但是,關(guān)于寄主植物識別MoSM1,并且如何激活下游抗病信號通路的機制研究還沒有報道。為了鑒定出水稻中能夠識別并結(jié)合MoSM1蛋白的受體蛋白,我們利用酵母雙雜交和蛋白質(zhì)免疫沉淀結(jié)合液相串聯(lián)質(zhì)譜實驗篩選與MoSM1的寄主受體蛋白。在酵母雙雜交實驗中,將含有pGBKT7-MoSM1的Y2HGold菌株與含有水稻的cDNA文庫的酵母Y187菌株進行mating。最后得到30個陽性克隆,并根據(jù)這些基因所編碼的蛋白的功能將范圍縮小到8個。但接下來的驗證互作實驗中,發(fā)現(xiàn)它們均不能檢測出互作信號,因此,酵母雙雜交實驗未能鑒定出與MoSM1互作的水稻蛋白。利用IP-MS技術(shù)篩選鑒定MoSM1的水稻受體蛋白。實驗中設(shè)置了三個實驗組及一個陰性對照普通型水稻XS11。提取水稻葉片總蛋白進行IP實驗。將所得到的蛋白進行質(zhì)譜分析。質(zhì)譜分析鑒定到膠條中含有220個蛋白,對這些蛋白進行GO功能分析,并依據(jù)蛋白功能、結(jié)構(gòu)域、亞細胞定位等指標(biāo)來初步篩選出約20個候選互作...
