臍帶間充質(zhì)干細(xì)胞對(duì)NB4白血病細(xì)胞基因組表達(dá)譜的影響

【摘要】:背景:間充質(zhì)干細(xì)胞是體內(nèi)微環(huán)境的重要組成部分且影響腫瘤細(xì)胞的多種生物學(xué)行為,間充質(zhì)干細(xì)胞的潛在臨床價(jià)值是近年來(lái)的研究熱點(diǎn)。目的:通過(guò)基因芯片方法分析臍帶間充質(zhì)干細(xì)胞對(duì)急性早幼粒細(xì)胞白血病細(xì)胞系NB4細(xì)胞基因組表達(dá)譜的影響。方法:體外建立臍帶間充質(zhì)干細(xì)胞與急性早幼粒細(xì)胞白血病細(xì)胞系NB4細(xì)胞共培養(yǎng)體系,檢測(cè)臍帶間充質(zhì)干細(xì)胞對(duì)NB4細(xì)胞增殖、凋亡和分化的影響,分別提取了NB4細(xì)胞單獨(dú)培養(yǎng)組和NB4細(xì)胞+臍帶間充質(zhì)干細(xì)胞共培養(yǎng)組NB4細(xì)胞的mRNA并將其反轉(zhuǎn)錄為cDNA,兩組分別標(biāo)記熒光,做成探針并與基因芯片雜交,檢測(cè)其熒光強(qiáng)度,分析兩組基因表達(dá)譜的差異。結(jié)果與結(jié)論:(1)臍帶間充質(zhì)干細(xì)胞促進(jìn)NB4細(xì)胞的增殖和分化并抑制其凋亡;(2)臍帶間充質(zhì)干細(xì)胞作用后的NB4細(xì)胞的基因表達(dá)譜中有530個(gè)基因上調(diào)和53個(gè)基因下調(diào),與基因相關(guān)的功能和信號(hào)通路也在一定程度上受到了調(diào)節(jié);(3)結(jié)果表明,臍帶間充質(zhì)干細(xì)胞通過(guò)改變腫瘤細(xì)胞內(nèi)大量基因表達(dá)、基因功能及多種信號(hào)通路影響NB4細(xì)胞的生物學(xué)行為。
[Abstract]:Background: mesenchymal stem cells are an important part in the micro environment and the influence of various biological behavior of tumor cells, mesenchymal stem cells and its potential clinical value is a hot research topic in recent years. Objective: to analyze the umbilical cord mesenchymal stem cells on acute promyelocytic leukemia cells NB4 gene expression profiles affected by the gene chip method. Methods: human umbilical cord mesenchymal stem cells and acute promyelocytic leukemia cell line NB4 cells co culture system in vitro, detection of umbilical cord mesenchymal stem cells on NB4 cell proliferation, apoptosis and differentiation, were extracted and NB4 + cells in human umbilical cord mesenchymal stem cells co cultured NB4 cells mRNA and reverse transcribed into cDNA NB4 cells were cultured alone, two groups were labeled with a fluorescent probe, and made hybridization and gene chip, detecting the fluorescence intensity, analysis of two groups of gene expression difference ISO. Results and conclusion: (1) human umbilical cord mesenchymal stem cells promote the proliferation and differentiation of NB4 cells and inhibit their apoptosis; (2) human umbilical cord mesenchymal stem cells after NB4 cell gene expression profiles of 530 genes up-regulated and 53 genes were down regulated, and the signal pathway and gene function related to a certain extent by the regulation; (3) the results showed that the umbilical cord mesenchymal stem cells by altering the expression of a large number of genes in tumor cells, gene function and signaling pathways of biological behavior of NB4 cells.
