利用交流阻抗傳感技術(shù)實時監(jiān)測納米溝槽上皮膚細(xì)胞的定向行為

【摘要】:細(xì)胞定向行為在皮膚、骨骼等修復(fù)和再生中起著非常重要的作用.本文首次采用交流阻抗傳感技術(shù)實時監(jiān)測人真皮成纖維細(xì)胞(HFF)和人永生化表皮細(xì)胞(Ha Ca T)在納米溝槽(寬度:200 nm,周期:400 nm,深度:70 nm)上的定向行為.結(jié)果表明,HFF細(xì)胞在納米溝槽上先進行定向排列,再發(fā)生胞體的延長;Ha Ca T細(xì)胞無定向行為的產(chǎn)生,其粘附和鋪展得到了延緩.與平面電極相比,HFF細(xì)胞在納米溝槽上產(chǎn)生的交流阻抗信號(NI值)更大,前期定向排列比后期胞體延長引起的NI值變化更顯著,且NI值與定向排列的細(xì)胞百分比之間存在著良好的線性關(guān)系;Ha Ca T細(xì)胞在納米溝槽上的NI值更小,且粘附比鋪展過程對NI值變化的影響更大.本文的研究將為復(fù)合型細(xì)胞傳感器的發(fā)展提供思路和支持.
[Abstract]:Cell orientation behavior in the skin. Bone repair and regeneration play a very important role. In this paper, the first real time monitoring of human dermal fibroblasts (HFFs) and human immortalized epidermal cells (Ha-Ca T) using AC impedance sensing technique was carried out. In nanogrooves (width: 200 nm). The results showed that the HFF cells were orientated first and then prolonged on the nanogutaneous grooves at the depth of 70 nm, and the cell cycle was 400 nm at the depth of 70 nms. the results showed that the HFF cells were orientated first and then the cell bodies were lengthened. The adhesion and spreading of Ha Ca T cells were delayed, and the AC impedance signal (NI value) of HFF cells on nano-grooves was larger than that of planar electrodes. The change of NI value in the early stage was more significant than that in the late stage, and there was a good linear relationship between the NI value and the percentage of the cells in the orientation arrangement. The NI value of Ha Ca T cells on nano-grooves is smaller, and the effect of adhesion ratio spreading on NI values is greater. The research in this paper will provide ideas and support for the development of complex cell sensors.
