桿狀病毒—腺相關(guān)病毒載體基因修飾骨髓間充質(zhì)干細胞靶向喉咽癌放射性 131 I治療研究

目的:喉咽癌是頭頸部常見的惡性腫瘤,預(yù)后極差。基于干細胞移植的基因療法有望改善治療效果。本研究中,我們明確人骨髓間充質(zhì)干細胞(BMSCs)是否具有潛在喉咽癌歸巢能力,探索桿狀病毒-腺相關(guān)病毒載體(BV-AAV)介導(dǎo)碘化鈉同向轉(zhuǎn)運體(NIS)基因修飾BMSCs靶向喉咽癌放射性131I治療的應(yīng)用價值。方法:1.構(gòu)建含有Luc-P2A-eGFP或NIS序列,側(cè)翼插入AAV來源的反向末端重復(fù)序列(ITR)的BV-AAV載體(Bac-CMV-Luc-P2A-eGFP-ITR和Bac-CMV-NIS-ITR)。2.從感染效率,細胞毒性,多基因的正確表達這三個方面評價了BV-AAV在BMSCs中的應(yīng)用價值。3.體外通過Transwell遷移試驗;體內(nèi)通過生物發(fā)光成像(BLI)和微單光子發(fā)射計算機斷層掃描成像技術(shù)(micro-SPET/CT)示蹤BMSCs體內(nèi)生物學(xué)分布和腫瘤趨化性。4.利用18F-FDG micro-PET/CT評估治療后腫瘤糖代謝情況;從腫瘤體積變化和腫瘤組織中細胞增殖標(biāo)志物表達來評估治療后腫瘤增殖情況。結(jié)果:1.MOI=400,Bac-...
