芒果GGPPS小亞基基因的克

【摘要】:香葉基焦磷酸(GPP)是香味成分單萜的前體物質(zhì),而香味是芒果重要的品質(zhì)性狀。本研究克隆了芒果中GPP合成關(guān)鍵酶基因,即香葉基香葉基焦磷酸合成酶小亞基基因(MinGGPPSssu),并開展了系統(tǒng)進化分析和表達分析。從芒果栽培品種貴妃芒的新鮮葉片中提取DNA和RNA,根據(jù)轉(zhuǎn)錄組測序拼接結(jié)果設(shè)計引物擴增得到Min GGPPSssu全長約1 kb的c DNA序列。通過測序、多重比對和聚類分析,證實該片段屬于GGPPSssu的全長基因,且為最大的ORF。與近緣物種苦楝GGPPS1(KM108317)氨基酸序列同源性為83%。蛋白質(zhì)序列分析表明該序列含有1個DDx_(2-4)D保守基元和一個Cxxx C基元。我們選取了擬南芥中12個GGPPS基因、2個FPPS基因、2個SPPS基因、一個GPS基因及其他物種的相關(guān)基因,通過聚類分析,將Min GGPPSssu聚類到香葉基香葉基焦磷酸合成酶小亞基所屬的枝。表達分析表明,該基因在葉片中表達量最高,莖部表達量較低。果皮和果肉在芒果成熟前后表達量相似,但是在果皮中的表達量約為果肉表達量的3倍,推測其與果皮及葉片香味調(diào)控相關(guān)。
[Abstract]:(GPP) is the precursor of monoterpenes, and aroma is an important quality trait of mango. In this study, the key enzyme gene of GPP synthesis in mango, the small subunit gene (MinGGPPSssu), of fragrant leaf pyrophosphate synthase, was cloned and the systematic evolution and expression analysis were carried out. DNA and RNA, were extracted from fresh leaves of mango variety GuifeiMang. According to the results of sequencing and splicing of transcriptional group, primers were designed to amplify the c DNA sequence with a total length of 1 kb of Min GGPPSssu. Through sequencing, multiple alignment and cluster analysis, it was confirmed that the fragment belonged to the full-length gene of GGPPSssu and was the largest ORF..
