大豆基因組中OSCA基因家族的進(jìn)化和表達(dá)分析

【摘要】:OSCA是鈣通透性陽(yáng)離子通道蛋白,在高滲脅迫感應(yīng)中發(fā)揮重要的作用。本研究利用生物信息,篩選并鑒定出21個(gè)大豆OSCA家族成員。通過染色體定位及同源性分析表明,OSCA家族成員分布在15條染色體上,且存在大量的復(fù)制現(xiàn)象;通過Ka/Ks分析表明大豆OSCA家族基因的進(jìn)化經(jīng)受純化選擇壓力。通過構(gòu)建系統(tǒng)進(jìn)化樹,將大豆OSCA家族分為2個(gè)亞家族,第一亞家族又分為三組(GroupⅠ~Ⅲ),且各組內(nèi)成員具有相似的內(nèi)含子-外顯子組成模式。保守性分析結(jié)果表明,大豆OSCA基因家族很保守,都包含3個(gè)功能結(jié)構(gòu)域(即Late exocytosis,Cytosolic domain of 10TM putative phosphate transporter和Calcium-dependent channel);進(jìn)一步組織表達(dá)分析結(jié)果表明,OSCA基因家族在不同組織的表達(dá)量不同。鹽堿脅迫表達(dá)模式分析表明,OSCA3.1上調(diào)表達(dá)變化倍數(shù)最大。進(jìn)一步的啟動(dòng)子分析結(jié)果表明,這些鹽堿脅迫應(yīng)答的基因啟動(dòng)子中存在著多種不同的脅迫響應(yīng)元件,其中干旱和ABA應(yīng)答相關(guān)的元件較多。以上結(jié)果為OSCA基因家族提供了新的有價(jià)值的信息,并為后續(xù)研究提供了重要參考。
[Abstract]:OSCA is a calcium permeable cationic channel protein, which plays an important role in hyperosmotic stress induction. In this study, 21 soybean OSCA family members were screened and identified by biological information. The results of chromosome localization and homology analysis showed that the members of OSCA family were distributed on 15 chromosomes and there were a large number of replication phenomena.
